若干杂交稻亲本和抗源的遗传多样性分析

【摘要】 本研究应用SSR标记检测了12个抗源与53个骨干亲本间的多态性,初步建立了这些亲本的SSR数据库。所用79个分子标记均能在亲本间揭示多态性,其中7个与抗源紧密连锁的标记(PTA248、SRM231/RM206、RM206、MRG4766、FP1+FP2+RP/AP22和SRM24/AP22)可以作为抗病基闪Xa-21、Xa-23、Pi-k~h、Pi-1、Pi-9和Pi-2前景选择的首选标记;47个SSR标记具有高PIC值(>0.5)且较均匀地分布在整个基因组中,推荐作为遗传多样性检测的首选标记。以SSR标记和18个表型性状为指标,分别对65个供试材料进行了聚类分析。利用SSR分子标记将供试材料划分为籼、粳两大类群,其中,籼稻群分为5个亚群,包含了恢复系亚群和保持系亚群;粳稻群分为4个亚群,包含了北方与南方粳稻亚群。而所检测12个抗源有10个抗源分布在籼稻群(恢复系群4个,保持系群4个,2个细条病抗源单独1个群),另外2个分布在粳稻群。表型聚类的划分结果与SSR标记聚类结果基本相同。
【Abstract】 The polymorphisms between the main parents and anti-sources were surveyed by molecular markers,and initially established the parents’ SSR database.79 pairs of primers can be used to reveal the polymorphism between the parents,in which seven primers,PTA248,SRM231/RM206,RM206,MRG4766,FP1+FP2+RP/AP22 and SRM24/AP22,that are closely linked with resistant genes,Xa-21,Xa-23,Pi-k~h,Pi-1,Pi-9 and Pi-2,are recommended as the preferred choice markers in foreground selection,and 47 primers,with high value of PIC(>0.5) and more evenly distributed throughout the genome,are recommended for analysis of genetic diversity for breeders.Cluster analysis based on phenotypic characters and SSR molecular markers were compared to study the diversity of 65 varieties.SSR cluster analysis classified the entries into two groups,Indica and Japonica.The Indica group consisted of 5 sub-groups,including the maintainer and restorer line subsets.The Japonica line group consisted of 4 sub-groups,including the north and the south Japonica subsets.Among 12 anti-sources,4 varieties were contained in restorer line subset,4 varieties were contained in the maintainer subset,2 varieties with bacteria leaf steak resistance were clustered together in an Indica subset,and 2 varieties were contained in the Japonica group.The phenotypic character cluster analysis confirmed the result based on the SSR markers clustering.
