The relationship between Thinopyrum intermedium and its four relatives revealed by comparative transcriptomics

【摘要】 In the natural evolution process,great changes of sequences have taken place between Thinopyrum intermedium(2 n=6 x=42,E~eE~eE~bE~bStSt/JJJ~SJ~SStSt) and its relatives Th.elongatum(2 n=2 x=14,E~eE~e),Th.bessarabicum(2 n=2 x=14,E~bE~b),Pseudoroegneria strigosa(2 n=2 x=14,StSt) and Th.Ponticum(2 n=10 x=70,E~eE~eE~bE~bE~xE~xStStStSt).In order to understand the evolutionary patterns of Th.intermedium and four relatives,the difference in their RNA-seq data was detected.RNA-seq was used to search both homologous genes and divergent genes from Th.intermedium and the four relatives.The evolutionary relationship and naturally selected genes of them were analyzed by the method of comparative transcriptomics.In total,41 divergent genes,accounts for only 0.81%,were found in orthologous genes.Functions of these genes were related to biologicalprocess and cellular component.Bayesian phylogenetic tree of Th.intermedium and its four relatives showed the relationship between Th.intermedium and Ps.strigosa is closer than that with Th.elongatum and Th.bessarabicum.The formation time of Th.intermedium is the latest among five plants.Additionally,the relationship between Th.elongatum and Th.bessarabicum is also cbser.The results showed that one of original parents of Th.intermedium is Ps.strigosa,and Th.elongatum and Th.bessarabicum are two putative genome donors.Under natural selection,the evolution of these plants tends to conservative rather than divergence.
