超高效液相色谱-串联质谱法测定婴儿尿液中2种雄激素
Determination of 2 Kinds of Androgen in Infant Urine by Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry

【摘要】 目的:建立尿液中睾酮和雄烯二酮的液相质谱-串联质谱检测法。方法:尿液经酶解后,使用乙酸乙酯萃取,采用Hypersil GOLDTMaQ C18色谱柱(100×2.1 mm,1.9μm)进行分离,正离子条件下三重四级杆串联质谱多反应监测模式进行检测,内标法定量。结果:2种雄激素的相关系数均大于0.99,定量限均为0.05μg/L,平均回收率范围为74.7-112.0%,相对标准偏差(RSD%)为4.6-15.7%。结论:该方法准确可靠,可以应用于婴儿尿液中睾酮和雄烯二酮的准确测定。
【Abstract】 Objective:To develop a method for determination of testosterone and androstendione in infant urine by ultra high performance liquid chromatography- tandem mass spectrometry.Methods:The analytes were extracted with ethyl acetate after enzymatic hydrolysis.The separation was carried out on a Hypersil GOLDTM aQ C18column(100 ×2.1 mm,1.9 μm) with methanol and water as mobile phase.The quantification was accomplished by internal standard method.Results:The calibration curves showed good linearity for both analytes with detection limits of 0.05 μg/L.The spiked recoveries in blank matrix ranged from 74.7-112.0%with RSDs less than 20%.Conclusion:The method was accurate and reliable,and was suitable for the determination of testosterone and androstendione in infant urine.
