保肺定喘汤对COPD气道重塑大鼠MMP-9、VEGF水平的影响

【摘要】 目的探讨保肺定喘汤对COPD气道重塑大鼠MMP-9、VEGF水平的影响。方法通过烟熏加气道滴入脂多糖(LPS)双重刺激法建立COPD气道重塑大鼠模型,观察保肺定喘汤干预治疗后各组大鼠一般情况、肺组织HE染色与Masson染色病理变化,使用ELISA法检测MMP-9、VEGF的水平。结果大鼠肺组织的病理改变显示COPD气道重塑大鼠模型建立成功且治疗前后病理改变有差异。ELISA法检测表明与正常组比较,模型组血清、肺泡灌洗液(BLAF)、肺组织中MMP-9、VEGF含量明显增高(P<0.01,P<0.05)。保肺定喘汤干预治疗后,与正常组相比,保肺定喘汤组血清、肺泡灌洗液、肺组织中MMP-9、VEGF含量仍较高,但较模型组MMP-9、VEGF含量降低明显(P<0.01,P<0.05)。结论保肺定喘汤可降低COPD气道重塑大鼠MMP-9、VEGF的含量,这可能与保肺定喘汤减缓COPD大鼠气道重塑的作用相关。

【Abstract】 Objective:Discussion Baofei Dingchuan Tang airway remodeling of COPD rat MMP-9,VEGF levels of influence.The method of smoking dropped by adding the airways lipopolysaccharide(LPS) stimulation double establish COPD airway remodeling in rats,observed after Baofei Dingchuantang intervention rats general,lung tissue HE staining and Masson staining pathological changes,using an ELISA assay MMP-9,VEGF levels.Results pathological changes of lung tissue in rats show that COPD airway remodeling in a rat model of pathological changes before and after the establishment of successful and treatment are different.ELISA assay showed that compared with the normal group,model group,serum and bronchoalveolar lavage fluid(BLAF),lung tissue MMP-9,VEGF were significantly increased(P <0.01,P <0.05).After Baofei Dingchuantang intervention,compared with the normal group,Baofei Dingchuantang serum,bronchoalveolar lavage fluid and lung tissue MMP-9,VEGF content is still high,but compared with the model group,MMP-9,VEGF decreased significantly(P <0.01,P <0.05).Conclusion Baofei Dingchuantang reduce airway remodeling in COPD rat MMP-9,VEGF content,which may slow down COPD Baofei Dingchuantang rat airway remodeling associated.
