纳米传感器在单细胞和疾病标志物检测中的应用

【摘要】 在生物和健康技术领域,能够实现生物流体时间分辨(time-resolved)和空间分辨(spatially resolved)的代谢物浓度测量的传感器(sensor)越来越受到重视,因为代谢物浓度检测传感器可以用于生物医药诊断和加强我们对于这些生物和疾病过程的了解。[1]比如细胞中H+(pH)的变化在细胞的分裂增殖、凋亡、多药抗药性、离子传输、内吞、和肌肉的收缩起到很重要作用。[2]除此之外监控细胞中pH的变化可以用于研究细胞内化途径(cellular internalization pathways)。针对目前纳米传感器存在的问题,通过在纳米微球上引入两种检测范围互补的荧光分子实现大量程的pH纳米传感器。,荧光比率(ratiometric)纳米传感器其敏感和参比荧光分子比例无法精确固定,导致不同批次纳米传感器的性能(标准曲线)不一致。以及不同批次性能(标准曲线)一致的纳米传感器,我们采用点击化学,高效合成多荧光组分比例精确固定的复合分子,制备批次问性能稳定、标准曲线一致的纳米荧光传感器。

【Abstract】 In the field of biology and health technology,sensors that are able to perform measurements of metabolite concentrations in biological fluids are in great demand.Nanosensors based on fluorescent ratiometric method for metabolite concentrations analysis were developed rapidly.We prepared pH nanosensor with extended measurement range by adding two pH indicator fluorescence dyes.It was found that standard curve of prepared nanosensors varied in different batches,which was caused by the minor change sensitive dye/reference dye ratio.Here,click chemistry is chosen to produce multi fluorescent blocks molecule at a high yield.Then the molecule was conjugated to nanoparticles to give nanosensors which have robust and constant standard curve in different batches.Fluorescent nanoparticles used for biomarker MYO test are also discussed here.
