Detection of CA15-3 using CdSe Quantum Dots@antibody based on ELISA

【摘要】 乳腺癌是迄今为止危害女性健康最常见的肿瘤疾病。糖连抗原CA15-3作为此类疾病中最重要的标识物,在健康人体内的此抗原的含量一般在30 U/mL以下,而30-50%的乳腺癌患者该指标明显升高,医学上将检测CA15-3的浓度视为监测乳腺癌的重要手段。因此,开发能够快速、灵敏检测CA15-3浓度的方法,对乳腺癌前期诊断、治疗以及术后的检测都具有极其重要的要义。本文借助酶联免疫吸附法(ELISA),将可对CA15-3识别的抗体1包覆在96孔板上,将可对CA15-3识别的抗体2通过共价键(生成-CONH-键)连接上CdSe量子点(发射峰:618 nm)表面,然后在血清溶液中可以对CA15-3进行定量检测(Figure 1A)。通过对样本稀释,可实现在0.2~18U/mL浓度范围内对CA15-3的线性检测(Figure 1B),其检测限可低至0.21U/mL(S/N=3)。
【Abstract】 Breast cancer is common malignant tumors for female disease.CA15-3 is one of the most important breast tumor markers and the normal level is under 30 U/mL in human serum.The concentration of CA15-3 could provide direct information to monitor the patients’ postoperative status and it can also predict recurrence and metastasis of breast cancer.Therefore,rapid and sensitive detection of CA15-3 is very important.Owing to the significance of ELISA in diagnosis and therapy,the development of sensitive detection of CA15-3 based on this method is highly desirable.Herein,we presented an approach by combining CdSe quantum dots(QDs) with the specific antibody for detecting CA15-3(0.2~18U/mL) with ELISA technology,and the limit of detection is found low to 0.21 U/mL.
