无标记DNA微悬臂生物传感器对脱氧核糖核酸酶生物活性的检测与研究

【摘要】 脱氧核糖核酸酶(DNase I)是一种很重要的DNA内切酶,它能够切断单链和双链DNA的磷酸骨架,同时它也是一类很有用的生物标记物。之前有研究报道前列腺癌和全身性红斑狼疮病人抑制了体内DNase I的活性,而心肌梗死病人增强了体内DNase I的活性[1];因此对DNase I含量的检测及活性的研究尤为重要。为此我们构建了一种基于微悬臂阵列的无标记、灵敏的生物传感器。先将76个碱基的单链DNA修饰在检测悬臂梁上作为DNase I的识别分子,为避免非特异性吸附,用6-巯基-1-正己醇封闭参比悬臂和检测悬臂的其他位点[2]。当溶液中存在DNase I时,由于DNase I能切断DNA的磷酸骨架,使DNA分子链变短,导致悬臂表面的应力发生变化,产生响应信号。这种检测DNase I的方法对研究其他的生物酶也有很大的应用前景。

【Abstract】 Deoxyribonuclease Ⅰ(DNase Ⅰ) is an important enzyme that cleaves both double-stranded and single-stranded DNA at their phosphate backbone.Previous studies have shown that patients with prostate cancer and systemic lupus erythematosus exhibit reduced DNase Ⅰ activity,and patients with myocardial infarction exhibit increased DNase I activity[1].So it is important to investigate the activity of DNase Ⅰ.A label-free detection method of DNase I using single-stranded DNA reporter-based cantilever array sensor was developed.The sensing cantilevers were functionalized with 76-bp DNA,while the reference cantilevers were modified with 6-mercapto-l-hexanol[2] to eliminate the influence of nonspecific adsorption.The interaction between DNase Ⅰ and DNA induced changes of cantilever surface stress,which caused a differential deflection between the sensing and reference cantilever.The method for DNase Ⅰ detection may have great potential for investigating other enzyme.
