pH敏感的水溶性阳离子卟啉类G-四链体光学探针及其在pH传感和DNA分子逻辑门中的应用

【摘要】 设计并合成了一种水溶性的阳离子卟啉衍生物5,10,15,20-四-{4-[3-(1-甲基-1-哌啶基)丙氧基]苯基}卟啉(TMPipPrOPP),并在分子拥挤条件环境下考察了它对不同构型DNA的光学响应。酸性条件下,TMPipPrOPP通过π-π堆积和静电相协同的方式与G-四链体结合形成稳定的TMPipPrOPP/G-四链体复合物,并产生明显的紫外和荧光信号响应;而在中性和碱性条件下,TMPipPrOPP/G-四链体复合物被破坏,光学信号消失。该复合物的形成与破坏可通过反复调控pH可逆进行(图1)。对于单链、双链和i-motif结构的DNA而言,在pH=5~8.5范围内均不产生光学响应。利用以上发现,设计了比色和荧光双比率的pH传感平台,并构建了AND,NAND,OR,NOR,INHIBIT和IMPLICATION等六种光学双输出的DNA分子逻辑门。

【Abstract】 A water soluble cationic porphyrin derivative 5,10,15,20-tetra-{4-[3-(l-methyl-l-piperidinyl)propoxy]phenyl}porphyrin tetraiodide(TMPipPrOPP) was synthesized and characterized.Under molecular crowding conditions,a stable TMPipPrOPP/G-quadruplex complex was formed in acidic pH range via the synergy between π-π stacking and electrostatic interactions.Alkalization of the system to neutral and basic pH ranges caused destruction of the complex due to reduced electrostatic interactions.The formation and destruction of the TMPipPrOPP/G-quadruplex complex was reversibly adjusted by pH,accompanied by repeated on-off switching of the colorimetric and fluorescent responses of TMPipPrOPP to G-quadruplexes(Fig.1).In comparison,no optical responses were observed for single-stranded,duplex and i-motif DNAs in the tested pH range(5.0~8.5).Based on above observation,colorimetric and fluorescent sensors for ratiometric pH-sensing were designed and DNA-based dual-output all-optical molecular logic gates that can perform six basic logic operations,including AND,NAND,OR,NOR,INHIBIT and IMPLICATION operations were constructed.
