柞蚕微孢子虫UGPase基因的克隆及序列分析

【摘要】 对柞蚕微孢子虫cDNA文库的EST序列进行测序分析,其中一条EST的重复数达到了94次,在测定的626个高质量EST中所占比例达到了15%。经过Blast比对分析,发现该基因与其它物种的UTP-葡糖-1-磷酸尿苷酰转移酶(UTP-glucose-1-phosphate uridylyltransferase),也称为尿苷二磷酸葡萄糖焦磷酸化酶(UDP-glucose pyrophosphorylase,UGPase)具有很高的相似性。通过进一步的基因克隆和序列测定,得到了柞蚕微孢子虫的基因全长,命名为NpUGPase(NCBI登录号:KJ210756)。该基因ORF长度为1 440 bp,编码479个氨基酸,预测的蛋白质分子量质量为54.3 kD,等电点6.24。通过与其它物种的同源序列之间比对,采用邻位相连法(Neighbour-Joining)构建系统发育树,结果显示柞蚕微孢子虫UGPase与其它真菌类的UGPase蛋白亲缘关系较近。该基因在真核生物和原核生物中存在一定差异。对柞蚕微孢子虫UGPase基因的研究,为更深入的探讨柞蚕微孢子虫的糖代谢途径、特异性降低糖类的合成、减少微孢子虫的感染率奠定基础。
【Abstract】 After analyzing ESTs from the c DNA library of Nosema pernyi, we found the number of replication one EST has reached for 94 times which account for 15% in 626 high quality ESTs we have sequenced. Through Blast analysis, this gene was homologous to UTP-glucose-1-phosphate uridylyltransferase(also named UDP-glucose pyrophosphorylase,UGPase) with other species. UGPase gene has cloned and named Np UGPase(Gen Bank accession number: KJ210756). This gene contained an ORF of 1 440 bp encoding a protein of 479 amino acids. The predicted molecular weight and isoelectric point of this protein was 54.3 k D and 6.24. Compared with other homology sequences of other species, a phylogenetic tree has constructed using Neighbour-Joining method, and the result showed that UGPase in N. pernyi has a closer relationship with other fungi UGPase proteins. This gene in prokaryotic is different with eukaryotic pyrophosphorylases. Cloning and research Np UGPase gene can provide a foundation for further study on glycometabolism, and reduce the infection rate through specific decrease carbohydrate synthesis in N. pernyi.
