【摘要】 目的:从胃肠动力学方面探讨硝菔通结方治疗功能性便秘的作用机制。方法:雄性SD大鼠50只随机分成正常组,模型组,硝菔通结方小、中、大剂量组,Powerlab检测大鼠在体结肠肌电的变化;ELISA法检测血清中胃动素(MTL)、胃泌素(GAS)的含量;Q-PCR检测大鼠结肠组织中VIPmRNA的表达变化。结果:硝菔通结方大剂量组大鼠结肠肌电频率升高、振幅降低(P<0.01),中、小剂量组频率增加(P<0.05);硝菔通结方大剂量组大鼠血清MTL、GAS含量显著性升高(P<0.01),中剂量组大鼠血清中MTL含量升高较明显(P<0.05);硝菔通结方大、中剂量组大鼠结肠组织VIPmRNA含量显著升高(P<0.05)。结论:硝菔通结方能够调节大鼠在体结肠肌电慢波频率和振幅,促进大鼠胃肠道的正向运动;提高大鼠血清中MTL、GAS的含量,提高结肠组织中VIP的表达,调节结肠过度的分节运动,增强胃肠动力,这可能是该方治疗功能性便秘的机制之一。更多还原

【Abstract】 Objective:Discussed from the aspects of gastrointestinal dynamics Xiaofu Tongjie Formula and mechanism of the treatment of functional constipation.Methods:Building rats randomly divided into model group,blank control group,functional constipation Xiaofu Tongjie Formula small,medium and large dose groups,Powerlab detect colon of rats in vivo electromyography changes;ELISA method to detect serum gastric dynamic element(MTL),the content of gastric secrete element(GAS);Q-PCR detection of VIPmRNA expression in rat colon tissue changes.Results:Xiaofu Tongjie Formula and the high-dose group rats colon elevated electrical frequency,amplitude,lower(P<0.01),increased frequency of medium and small dose group(P<0.05);Xiaofu Tongjie Formula and the high-dose group rats serum MTL,GAS levels increased significantly(P<0.01),medium dose group of rats serum MTL content increased obviously(P<0.05);Xiaofu Tongjie Formula square big,middle dose group rats colon tissue VIPmRNA content increased significantly(P<0.05).Conclusion:Xiaofu Tongjie Formula can adjust the colon of rats in vivo electrical slow wave frequency and amplitude,promote the positive movement rats gastrointestinal tract;Improve the rat serum MTL,GAS levels,improve the expression of VIP in colon tissue,adjust the segmental motion of colon is excessive,enhance the gastrointestinal dynamics,this may be one of the mechanisms of the party in the treatment of functional constipation.更多还原
