【摘要】 目的:观察四君子丸对Lewis肺癌小鼠肿瘤组织生长抑制作用及肿瘤细胞凋亡的影响。方法:在小鼠右侧腋窝皮下,接种浓度为1×10~7个/mL的小鼠肺癌Lewis细胞悬液0.1mL,模型建立成功后将小鼠分为4组:Lewis肺癌组、Lewis肺癌中药治疗组、Lewis肺癌中药预防组、Lewis肺癌顺铂组。Lewis肺癌中药治疗组:成瘤后灌胃四君子丸0.0936g/10g,每天1次,连续14d;Lewis肺癌中药预防组:造模开始灌胃四君子丸0.0936g/10g,每天1次,成瘤后加14d;Lewis肺癌顺铂组:腹腔注射顺铂0.05mg/10g,每周1次,持续2次。采用游标卡尺测量移植瘤直径,并计算肿瘤体积、相对肿瘤体积和肿瘤增殖率,采用TUNEL法测定肿瘤组织细胞凋亡情况。结果:与Lewis肺癌组比较,Lewis肺癌中药治疗组、Lewis肺癌中药预防组、Lewis肺癌顺铂组肿瘤体积、相对肿瘤体积显著降低(P<0.01);Lewis肺癌中药治疗组、Lewis肺癌中药预防组和Lewis肺癌顺铂组相对肿瘤增殖率分别为42.35%、45.47%和41.02%,且和Lewis肺癌组比较有显著性差异(P<0.01);TUNEL染色结果显示:Lewis肺癌组肿瘤组织有少量凋亡细胞,Lewis肺癌中药治疗组、Lewis肺癌中药预防组和Lewis肺癌顺铂组凋亡细胞数量显著增多,和Lewis肺癌组比较,Lewis肺癌中药治疗组、Lewis肺癌中药预防组和顺铂组凋亡指数显著升高(P<0.01)。结论:四君子丸能够促进Lewis肺癌小鼠模型肿瘤组织中细胞凋亡,进而抑制肿瘤增殖。更多还原

【Abstract】 Objective:to observe the effects of inhibition and apoptosis of four noble drugs on Lewis lung cancer in mouse.Methods:the mice were inoculated with Lewis lung cancer cells,the concentration of 0.1mL cells suspension is 1×10~7ind/mL witch was inoculated in right armpit subcutaneously of mouse model.The mouse model was divided into 4 groups:Lewis lung cancer group(control group),TCM treatment group,Chinese herb prevention group and PDD group.For TCM treatment group:four noble drugs 0.0936g/10 g QD by gastric perfusion after tumor formation,lasted for 14 days;for Chinese herb prevention group:four noble drugs 0.0936g/10 g QD by gastric perfusion at the beginning of creating the model,add 14 d after tumor formation;for PDD group:injected intraperitoneally with PDD 0.05mg/10 g once a week,lasted for two weeks.Diametering measurement of the xenograft with vernier caliper,and calculate the relative tumor volume and tumor appreciation rate.Detected the apoptosis of tumor tissues by TUNEL.Results:compared with control group,tumor volume and relative tumor volume of TCM treatment group,Chinese herb prevention group and PDD group significantly decreased(P<0.01),the relative tumor appreciation rate of TCM treatment group,Chinese herb prevention group and PDD,5mg/kg,group were 42.35%,45.47%and41.02%respectively,and the difference was significant compared with control group(P<0.05);Lewis lung cancer model group has a few apoptosis cells,apoptosis cells respectively increasing of TCM treatment group,Chinese herb prevention group,PDD group,by calculating the cell apoptotic index,the results showed that it’s respectively increasing of TCM treatment group,Chinese herb prevention group PDD group compared with Lewis lung cancer group(P<0.01).Conclusion:four noble drugs can reduce the tumor volume,increase the cell apoptotic index of tumor tissues,then to achieve the goal that inhibit tumor growth,decrease tumor appreciation rate.更多还原
