加味聚精食疗方对少、弱精子症大鼠EGF、EGFR睾丸内表达的影响

【摘要】 【目的】观察加味聚精食疗方对少、弱精子症大鼠表皮生长因子(EGF)、表皮生长因子受体(EGFR)睾丸内表达的影响。【方法】60只3月龄雄性SD大鼠采取区组随机化法分为6组:G1组、G2组、G3组、G4组、G5组、G6组,每组10只。G1组蒸馏水4g/kg灌胃52天;其余5组予雷公藤多甙(GTW)20mg/kg灌胃26天造模。造模结束后,G6组立即脱颈处死,以了解造模质量;G2组加味聚精食疗方6.3g/kg灌胃;G3组枸橼酸他莫昔芬片1.8mg/kg灌胃;G4组五子衍宗片0.4g/kg灌胃;G5组蒸馏水4g/kg灌胃,以上均每天一次,连续26天。治疗结束后处死各组大鼠,CASA技术检测大鼠附睾精子质量;对大鼠肝、肾、睾丸进行组织病理学检查;Envision两步法检测睾丸组织EGF、EGFR的表达。【结果】G6组附睾精子质量的各项指标均较G1组明显减低(P<0.05)。G2组附睾精子质量所有指标均明显优于G5、G6组(P<0.05)。G2组附睾精子密度明显优于G3、G4组(P<0.05)。G1、G2组附睾精子质量所有指标组间比较不具有统计学意义(P>0.05)。成熟大鼠睾丸组织EGF、EGFR阳性产物主要定位于睾丸间质细胞的胞浆和胞膜上,呈棕黄色的颗粒或者团块状,而在各级生精细胞、支持细胞及精子细胞上均未见表达。G1、G2组大鼠睾丸组织结构情况、曲细精管精子数量、睾丸组织EGF、EGFR表达水平明显优于G5、G6组(P<0.05)。【结论】加味聚精食疗方能显著上调大鼠睾丸间质细胞EGF、EGFR的表达水平,这可能是其治疗少、弱精子症的作用机制之一。

【Abstract】 Objective:To observe the effects of Jiawei jujing Decoction on Epidermal Growth Factor(EGF) and Epidermal Growth Factor Receptor(EGFR) expression in oligozoospermia or asthenospermia rats’ testes.Methods:60 three months male SD rats were randomized into G1,G2,G3,G4,G5,G6 groups(10 per group).Gl group received intragastric administration with distilled water(4g/kg)lavage for 52 days.While the other 5 groups were made modes by intragastric administration with Glucosidorum Tripterygll Totorum(GTW,20mg/kg) for 26 days.G6 group were executed immediately in order to know the quality of the mode.Then G2,G3,G4,G5 group received intragastric administration with JJD(6.3g/kg),Tamoxifen Citrate(1.8mg/kg),WYT(0.4g/kg),distilled water(4g/kg) for 26 days respectively.All rats were executed After the study.Rats epididymis sperm quality were tested by Computer-Assisted Sperm Analysis(CASA).And the liver,kidney,testicular received pathological examination.EGF,EGFR expression in adult rat testicular tissue were detected by Envision two-step.ResuIt:G1 group’s all indexs indicate a significant differences between G6 groups after the experiments(P<0.05).G2 group’s epididymis sperm quality was better than G5,G6 group,and G2 group’s epididymis sperm density was more dense than G3,G4 group(P<0.05).The difference of epididymis sperm quality between G1,G2 groups has no statistical significance after the experiments(P> 0.05).The EGF,EGFR positive products were tan-yellow particles,and crumb,and located in cytoplasm and cell membrane in mature rats testicular tissue’s leydig cells.All the spermatogenic cell,Sertoli cells or sperm cells did not express the EGF,EGFR at all.Gl,G2 group’s testicular tissue structure,sperm cell in seminiferous tubules,the EGF,EGFR expression level were more better than G5,G6 group(P<0.05).Conclusions:JJD can raise EGF,EGFR expression levels in rats’ testicular leydig cell significantly.And it might be one of the mechanism for JJD treating oligozoospermia or asthenospermia.
