大孔吸附树脂纯化葛根总黄酮的工艺研究

【摘要】 目的:研究大孔吸附树脂分离纯化葛根总黄酮的工艺,并为更深入研究脑脉通复方组分配伍奠定基础。方法:以总黄酮的吸附量和解吸率、出膏率为指标,对树脂类型、树脂径高比、上样浓度等工艺参数进行考察,确定最佳纯化工艺。结果:最佳纯化工艺为:采用AB-8大孔吸附树脂,径高比1:10,上样浓度为0.25 mg.mL-1,上样量为5 BV,水洗量为4 BV,洗脱溶剂为50%乙醇,洗脱用量为5 BV。结论:纯化工艺稳定可行。
【Abstract】 Objective:To study the purification technology of total flavonoids from Puerariae Radix with macroporous resin,and to make foundation for further studying the component combination of the effective parts from Naomaitong formula.Methods:With the absorption capacity,desorption rate of macroporous resin and the rate of extractum extracted as indexes,investigated the optimal kind of resins,diameter-height ratio of column,concentration of sample solution etal,to optimize the separation process.Results:the optimum purification conditions were as follows:AB-8 macroporous resin was chosen,diameter-height ratio of column was 1:10,concentration of sample solution was 0.25 mg.mL(-1),volume of sample solution was 5 BV,elution volume with water was 4 BV,elution volume with 50% ethanol was 5 BV.Conclusion:the optimium purfication technology was stable and feasible.
