补阳还五汤对实验性肺纤维化大鼠血清纤维化指标PCⅢ、Ⅳ-C含量的影响

【摘要】 目的观察补阳还五汤对博来霉素致肺纤维化大鼠血清纤维化指标PCⅢ、Ⅳ-C的影响。方法将144只健康雄性SD大鼠采用随机数字表法分成6组,假手术组、模型组、强的松组、补阳还五汤高、中、低组,每组各24只。除假手术组外,其余各组采用一次性气管滴注博莱霉素复制肺纤维化大鼠模型,假手术组气管内滴注等量生理盐水,造模第二天起,各给药组分别给予相应药物灌胃,于造模后第7d、14d、28d分3批处死动物,每次每组8只。分离血清,采用放免法检测各组大鼠血清纤维化指标PCⅢ、Ⅳ-C的含量。结果:①与假手术组相比,其它各组大鼠血清PCⅢ、Ⅳ-C的含量均逐渐升高,以模型组最为明显(p<0.01);②与模型组相比,各给药组大鼠血清PCⅢ、Ⅳ-C含量均有不同程度降低(p<0.01);③与强的松对照组相比,补阳还五汤中剂量组第14d、28d以及补阳还五汤低剂量组第28d时,PCⅢ、Ⅳ-C含量下降明显(p<0.05)。结论经补阳还五汤治疗后的模型大鼠血清纤维化指标含量有所下降,提示补阳还五汤可以有效抑制病肺组织中胶原纤维的增殖以及细胞外基质的沉积,起到较好的抗肺纤维化作用,其具体机制有待进一步研究。

【Abstract】 Objective To investigate the effects of Buyanghuanwu decoction on the expression of PCIII、IV-C in blood serum of pulmonary fibrosis rats induced by bleomycin(BLM).Methods 90 healthy male SD rats were randomly divided into six groups:sham operation group,model group,prednisone group,Buyanghuanwu decoction high- dose,middle- dose,low-dose groups,24 rats in each group.Pulmonary fibrosis model was established by injected with BLM by tracheal intubation,sham operation group was intratracheally instilled with equal NS,then every rats was raised drugs through a gastric tube one times a day.8 rats in each group were killed respectively on days 7,14,28 after intratracheal instillation,tested the contents of PCIII、IV-C in blood serum of experimental rats by radioimmanoassay(RIA) method.Results① Compared with the sham operation group,the contents of PCIII、IV-C in blood serum of other groups were gradually increased,and that of the model group was the most obvious(p<0.01);②Each medicated group had different decreases in the contents of PCIII、IV-C in blood serum by comparison with model group(p<0.01);③Compared with the prednisone group,the contents of PCIII、IV-C of Buyanghuanwu decoction middle-dose group obviously decreased on the 14th and 28th day(p<0.05),as well as that of Buyanghuanwu decoction low-dose group on the 28th day(p<0.05).Conclusion Buyanghuanwu decoction could effectively reduce the contents of PCIII、IV-C in blood serum of experimental rats by BLM,which proved that buyanghuanwu decoction could effectively control the reproduction of collagen fibers and excess deposition of ECM in lung tissue of PF,and played an anti-f ibrosis effect,the specific mechanism of that were expecting for further study.
