【摘要】 目的:筛选并验证与老龄肾阳虚证体质相关的差异表达基因。方法:采用SAM 3.1软件分析前期所获老龄肾阳虚差异基因表达谱,从差异表达基因谱中筛选出8个基因,并从差异表达基因功能、分子功能分类、分子通路等层面分析探讨老龄肾阳盛衰基因调控模式,通过调节Delta值,使得假阳性率为0,获得3个基因,选取10例肾阳虚证患者和10名正常对照者,采用半定量RT-PCR检测其血液中这3个基因的表达情况,验证与老龄肾阳虚证相关的这3条差异表达基因。结果:这3个基因在10例老龄肾阳虚证患者和10名正常对照者间的表达水平存在明显差异。结论:老龄肾阳虚证体质的相关基因涉及到衰老、肿瘤发生、生殖系统疾病等,集中体现了肾阳虚衰对本脏功能维持作用的减弱及脑肾关系。更多还原

【Abstract】 Objective:To filter and verify the relevance of the differentially expressed genes of aged kidney-yang deficiency syndrome constitution.Methods:From the earlier gene expression profile chip,8 genes in the differentially expressed genes were filtered and the gene regulation mode of kidney-yang deficiency from differentially expressed genes were studied,gene functional classification and molecular pathway by SAM 3.1.After delta regulation and false positive rate of 0 control,3 genes were filtered in the differentially expressed genes.10 elderly patients with kidney-yang deficiency and 10 healthy people were selected,and 3 genes’ expression were detected in the blood by the semi-quantitative skill of the RT-PCR,and the differentially expressed genes which correlated with the aged kidney-yang deficiency syndrome were verified.Results:There were 3 genes that had significant differences in expression between the 10 elderly patients with kidney-yang deficiency and 10 healthy people.Conclusion:The related genes of the aged kidney-yang deficiency syndrome was related to senility,tumor and reproductive system disease.It reflected that the function of kidney became weak from yang-deficiency and reflected the relationship between brain and kidney.更多还原
