【摘要】 目的:研究中药复方健脾解毒方对裸鼠人肠癌血管新生的抑制作用。方法:建立人肠癌COLO-205细胞裸鼠皮下移植瘤模型,随机分为5组:对照组（生理盐水组）,健脾解毒方低、中、高剂量组(250,500,1000 mg.kg^-1·d^-1)和阿霉素组(2mg.kg^-1.d^-1)。给药第21d,各组均处死荷瘤鼠,测量肿瘤大小及质量。免疫组化法检测瘤体的MVD和β-catenin表达;ELISA测定血清中VEGF的表达。结果:健脾解毒方低、中、高剂量组的瘤重抑制率分别为33.33%,36.54%,44.04%。免疫组化结果显示,对照组、健脾解毒方低、中、高剂量组瘤体MVD计数分别为63.67±3.21,50.33±4.51,47.00±3.00,34.67±4.51个,与对照组比较差异显著（P<0.01）。ELISA结果显示,健脾解毒方低、中、高剂量组血清VEGF表达与对照组相比显著下调（P<0.01）。健脾解毒方能够下调瘤体中β-catenin蛋白表达,并且呈一定剂量依赖效应。结论:健脾解毒方对人肠癌皮下移植瘤生长和血管新生具有一定抑制作用,其机制可能是通过Wnt/β-catenin信号通路下调VEGF表达有关。更多还原

【Abstract】 Objective:To study effects of Jianpi Jiedu Recipeto hypodermatic transplanted tumor icrangium growing in colorectal cancer nude mice.Method:Colorectal cancer models in nude mice bear human COLO-205 cells were established,and then were randomly divided into 5groups:Model group（NS control group）,Jianpi Jiedu Recipelow-dose,mid-dose,high-dosegroup(250,500,1000 mg·kg^-1·d^-1),ADRgroup(2 mg·kg^-1·d^-1).All group of nude mice were sacrificed at the last time treatment for 21 days,the size and weight of tumor weremeasured.The MVD and COX-2 was tested by Immunohistochemistry;the VEGFprotein expression in mice serum was assayed by ELISA.Results:The inhibition rate of tumor in each group were respectively33.33%,36.54%,44.04%in low,mid,and high-dose Jianpi Jiedu Recipe.The MVD in Model group and low,mid,high-dose Jianpi Jiedu Recipe were respectively 63.67±3.21,50.33±4.51,47.00±3.00,34.67±4.51,which was obviously decreased compared with Model group（P<0.01）.The VEGF in low,mid,high-dose Jianpi Jiedu Recipewas obviously decreased compared with Model group（P<0.01）.Jianpi Jiedu Recipe dose-dependently reduced the level of β-catenin in mice tumor.Conclusion:Jianpi Jiedu Recipe could inhibited the growth of colon carcinoma implant tumor of nude mice and MVD,which may be due to blocking Wnt/p-catenin signal pathway and reduced VEGF expression.更多还原
