【摘要】 目的:探讨健脾解毒方(Jianpijiedufang,JPJDF)对幽门螺杆菌(Helicobacter pylori,H.pylori)诱导胃癌细胞血管生长因子(Vascular endothelial growth factor,VEGF)、血管生成素-2(Angiopoietin-2,Ang-2)以及成纤维细胞生长因子(basic fibroblast growth fator,bFGF)表达的影响,揭示H.pylori感染促进胃癌生长、转移的部分机制。方法:采用ELISA法检测H.pylori标准株NCTC11637感染对人胃癌MKN45细胞VEGF、Ang-2和bFGF蛋白表达的影响。结果:H.pylori感染人胃癌MKN45细胞后VEGF、Ang-2和bFGF蛋白的表达明显上调,健脾解毒方可抑制三种蛋白的表达,并呈一定的剂量依赖效应。结论:H.pylor感染诱导胃癌细胞VEGF、Ang-2和bFGF蛋白的表达,这可能是H.pylori感染促进胃癌生长、转移的重要机制之一,健脾解毒方可抑制三种蛋白的表达,可能是其防治幽门螺杆菌相关胃癌的重要机制之一。更多还原

【Abstract】 Purpose:To investigate the effect of Jianpijiedufang(JPJDF) on Vascular endothelial growth factor(VEGF),Angiopoietin-2(Ang-2) and basic fibroblast growth fator(bFGF) expression in Helicoloacter pyloror-induced human gastric cancer cells,and reveal part of the mechanism of gastric cancer growth and metastasis promoted by H.pylori infection.Methods:The expression of VEGF,Ang-2 and bFGF protein in human gastric epithelial cell infected by standard H.pyloriNCTC11637 was evaluated by enzyme-linked immuno sorbent assay(ELISA).R esals:H.pylorisignificantly stimulated the expression of VEGF,Ang-2 and bFGF protein in MKN45 cell line(P<0.01),compared with MKN45 cell.PJDF could down-regulate VEGF,Ang-2 and bFGF protein expression induced by H.pylorin a dose-dependent manner compared with the H.py]ariinfection group.Conclusion:H.pylorican induce the expression of VEGF,Ang-2 and bFGF in human gastric epithelial cell,which might be one of the important mechanisms of gastric cancer growth and metastasis promoted by H.pyloriinfection.PJDF inhibitedH p-induced the expressions of VEGF,Ang-2 and bFGF,which may be one of its mechanisms in prevention and treatment ofH p-induced gastric cancer.更多还原
