心血瘀阻证动态演变过程大鼠模型的建立及方证验证评价的研究

【摘要】 目的:应用大鼠模型重现心血瘀阻证形成过程。方法:采用高脂饲料与冠状动脉左前降支结扎术结合的方法,分3阶段造模法。高脂饲料喂养至血脂增高模拟第一阶段"血瘀证前期";继续喂养至动脉粥样硬化,模拟第二阶段"亚血瘀证期"动物模型;在第二阶段成模的基础之上采用冠状动脉结扎术复制第三阶段"心血瘀阻证期"模型。成模后采用阳性药物、阴性药物干预法验证动物模型。观察每阶段大鼠表征,检测血液流变学、血脂指标。结果:①大鼠表征亚血瘀证组与血瘀证前组、心血瘀阻证两组比较差异具有显著性(P<0.01)。②血流变指标水平血瘀证前期组、亚血瘀证组、心血瘀阻证组水平次第增高,差异具有显著性(P<0.01)。③血脂指标水平以亚血瘀证组、心血瘀阻证组水平较血瘀证前期组高,差异具有显著性(P<0.01)。④方证验证阳性药物组血浆粘度、中切比较阴性药物组与模型组水平低,差异有显著性(P<0.05)。结论:通过三阶段造模法以及方证验证,建立了心血瘀阻证动态演变的实验动物模型。
【Abstract】 Objective:Rat model to reproduce the formation of blood stasis syndrome.Methods:A high—fat diet and the left anterior descending coronary artery ligation method of combining three stages modeling method.Fat diet to increased blood lipids simulated first stage " Early blood stasis syndrome ";continue feeding to atherosclerosis,animal models simulate the second stage " Asian blood stasis syndrome of ";On the basis of the second stage into the mold on top copy coronary artery ligation third stage " heart blood stasis period" model.After a positive mode using drugs,negative drug intervention method validation in animal models.Each rat was observed phase characterization,testing blood rheology,serum lipids.Results:CD characterization of sub- BSS group of rats with blood stasis syndrome former group,blood stasis syndrome difference was significant(P<0.01).(2) changes in blood flow pre- set target levels of blood stasis,sub- group of blood stasis,blood stasis syndrome group increased level of sequence,the difference was significant(P<0.01).(3) serum lipids levels in sub- BSS group,blood stasis stasis syndrome group compared with the high levels of early syndrome group,the difference was significant(P<0.01).(3) party card to verify positive drug plasma viscosity,low- cut negative for drugs compare with the model group level,the difference was statistically significant(P <0.05).Conclusion:Through a three-stage modeling method and party card verification,established animal model of blood stasis syndrome dynamic evolution.
