不同途径免疫小鼠重组乳酸乳球菌诱导的黏膜及系统免疫应答分析

【摘要】 重组乳酸乳球菌分别经鼻腔和口腔免疫小鼠后,免疫后的测定时间内鼻腔免疫方法刺激呼吸道和肠道广生s-IgA水平高于口服免疫方法,且肺泡冲洗液中s-IgA水平差异极显著(P>0.01),肠道黏膜s-IgA水平无显著差异(P>0.05);血清中IgG抗体水平两种免疫方法均无显著差异(P>0.05),但鼻腔免疫组抗体水平在最后一次免疫后7d达到最高,在整个测定时间内维持在高水平,口服免疫组抗体水平在最后一次免疫后21d达到最高水平,然后迅速下降;h 1型细胞因子IL-2和IFN-γ水平滴鼻免疫组高于口服免疫实验组,差异不显著(p>0.05),T h 2型细胞因子IL-4水平在各组之间差异不显著(p>0.05)。结果表明重组乳酸乳球菌分别经鼻腔及口腔免疫小鼠后,能诱导机体产生黏膜免疫系统的体液免疫及Th1型细胞免疫反应,且滴鼻免疫方法优于口服免疫方法,为传染病的预防控制提供了新的免疫策略。
【Abstract】 With the aim of determining the mucosal immune pathways of Recombinant Lactococcus lactis.we observed that Recombinant Lactococcus lactis administrated respectively intranasally and orally induced mucosal and systemic immune responses in mice.At the whole experimental session,the production of the s-IgA antibody in lung lavage fluid from the i.n immunized mice was higher significantly than that from the o.r immunized mice(P<0.01),the production of the s-IgA antibody in intestinal washes from the i.n immunized mice was higher than the o.r immunized mice((P<0.05).IgG anti-PRRSV was detected in serum,the level of special IgG from the i.n immunized mice was higher than the o.r immunized mice((P<0.05),however,at all time points.the i.n immunized mice get it’s maximum number on day 30,which lasted at least 4 weeks post-immunization:as well as the o.r immunized mice get it’s maximum number on day 44,following decreased rapidly;Interestingly,we investigated the activity of Th1-type cytokines IL-2,IFN-γ and Th2-type cytokines IL-4 in splenocytes on day 14 after the last immunization,the contents of cytokines IL-2 and IFN-γ in intranasal immunized mice was higher compared to that from the oral immunized mice((P>0.05),Notably,it is worthy to note that cytokines of IL-4 contents was not different to the all groups.All the results indicated that mice immunized by the recombinant L.lactis expressing ORF6 gene of PRRSV can significantly induce mucosal immune responses,humoral immunity responses and enhanced Th1 immunity response against the PRRSV,in contrast,Intranasal immunization is superior to oral immunization.
