表面等离子体共振法研究淀粉样β蛋白与金属离子的相互作用

【Abstract】 Amyloid β peptide plays a critical role in the pathogenesis of Alzheimer’s disease(AD). Transition metals, such as Cu2+ and Zn2+ are found in Aβ plaques, and they have been shown to affect Aβ aggregation. Here we investigated the interactions between metal ions and Aβ1-16 by a flow-injection surface plasmon resonance spectrometry(SPR). Our study revealed that both Zn2+ and Cu2+ promoted human Aβ1-16 conformation change but the Zn-induced confromational change is more stable. However, rat Aβ1-16 binds Zn2+ less avidly and hardly no conformational change, this may due to three point subsitutions, R5 G, Y10 F, and H13 R in rat Aβ1-16 which appear to alter the physicochemical properties of the peptide. The results demonstrate that SPR biosensor can be a sensitive and effective method to investigate the interaction mechanism between metal ions and amyloid β-peptide.
