杂交鳢胚胎发育及其基因组DNA甲基化修饰谱的研究

【摘要】 为简化杂交鳢亲本选育过程、提高杂交受精率及苗种成活率并解决杂交鳢苗种供应不足等问题。本研究以乌鳢、斑鳢为研究对象,通过人工催产授精获得乌鳢、斑鳢自交及正反交子代的胚胎,进行胚胎、仔鱼发育时序及各期形态变化的分析观察,确定自交子代与杂交子代胚胎发育过程的异同。通过定量PCR,比较分析dnmts基因的时序表达谱及胚胎基因组甲基化位点谱变化,以确定dnmts对自交、正反交子代早期发育甲基化模式的建立、维持及基因组甲基化位点多态性的调控作用,并确定早期胚胎基因组甲基化位点谱的变化与杂种优势形成的相关性,为进一步研究鱼类杂交育种提供新线索。
【Abstract】 To simplify the parent selection and improve the hybrids success, and increase the amount of hybrid off springs, in this study, the embryogenesis of the hybrids were observed and differences in development between parents inbreeds and hybrids were examined. Through RT-q PCR, the temporal expressions of dnmts were analyzed during embryogenesis. At same time we compared the dnmts transcript profiles and embryonic genomic DNA methylation patterns between the hybrid and its parent inbreeds, and investigated the relationships of gene expression, genome DNA methylation and phenotype of the hybrid, then find out the factors, regulated the super straits of hybrids. Thus the results would demonstrate the mechanisms behind the fish heterosis formation and provide new clues fish hybrid breeding in future.
