三斑海马脂溶性组分对脂多糖刺激小鼠单核巨噬细胞RAW264.7体外抗炎作用的研究

【摘要】 本研究探讨了海洋硬骨鱼三斑海马的脂溶性组分的抗炎活性。系统溶剂萃取海马粗粉95%乙醇回流提取物得到石油醚提取物,经硅胶柱层析,获得组分FrⅣ,经硅胶柱层析用不同溶剂洗脱分别获得中性脂,糖脂和磷脂组分,基本组分通过薄层层析和红外光谱分析。通过检测体外脂多糖刺激小鼠单核巨噬细胞RAW264.7炎症的促炎细胞因子分泌分析脂质的抗炎作用。结果表明三斑海马脂质与脂多糖刺激小鼠单核巨噬细胞RAW264.7一氧化氮,白细胞介素-1β,IL-6和肿瘤坏死因子-α产量有剂量依赖的体外抑制关系。在这些组分中,糖脂表现出较强抑制炎症细胞因子产生的能力,其和阳性对照品地塞米松(20μg/m L)的作用是相当的。我们的结果表明三斑海马脂质提取物也许对抗炎有效。
【Abstract】 The present study attempts to investigate the anti-inflammatory potential of the lipid soluble components isolated from three-spot seahorse which is rare marine bony fish. Petroleum ether(PE) extract was obtained from systematic solvent extraction after reflux extraction with 95% ethanol. FrⅣ was collected after silica gel column chromatography, and neutral lipids(NL), glycolipids(GL), phospholipids(PL) were separated from FrⅣ, respectively. Basic compositions were detected and analyzed via thin layer chromatography(TLC) and Fourier transform infrared spectroscopy(FTIR). Anti-inflammatory activities of total lipids(TL), isolated NL, GL, and PL were detected by secretion of pro-inflammatory cytokines induced by lipopolysaccharide(LPS) in murine monocyte macrophage RAW264.7 cells in vitro. The results revealed that lipids of seahorse showed a positive correlation with the in vitro suppression of the release of nitric oxide(NO), interleukin(IL)-6, IL-1β and tumor necrosis factor(TNF)-α potently in a dose dependent manner, showed cell compatibility. Among the fractions, GL(50μg/m L) showed the highest capacity to attenuate the generation of pro-inflammatory cytokines which was comparable to that of the positive drug dexamethasone(DX)(20μg/m L). Collectively, our findings indicated that the lipids from seahorse may be effective in the management of inflammation.
