嗜盐杆菌HSQAY1对中肋骨条藻的溶藻物质特性研究

【摘要】 为研究嗜盐杆菌溶藻菌株(Halobacillus sp.HSQAY1)产生的溶藻活性物质的特性,实验通过改变p H、温度、反复冻融条件研究其对中肋骨条藻溶藻活性的影响,并采取超滤、乙醇沉淀法等手段对溶藻物质进行了分离纯化研究。结果显示,该菌株产生胞外溶藻物质在酸性条件下(p H=3)丧失溶藻活性,而在p H 5~11时仍保持较高溶藻活性(溶藻效率87.0%~98.8%);随温度的升高溶藻活性虽略有下降趋势,但110℃高温处理后溶藻率仍可达83.2%;反复冻融对其溶藻活性没有影响。超滤结果表明该溶藻物质分子量>10 k Da;乙醇浸提相没有溶藻活性,沉淀相溶藻率为75.1%。

【Abstract】 The characteristics of algicidal substance produced by Halobacillus sp.(HSQAY1) under different p H, heat treatment, and repeated freeze-thaw cycles were investigated. The results showed that the algicidal substance was not significantly affected by alkali and effectively inhibited the growth of Skeletonema costatum, but lost the algicidal activity under stresses with acid treatment. In the heat treatment and the repeated freeze-thaw cycles treatment, the algicidal activity of the algicidal substance did not change significantly. The molecular weight of algicidal substance was > 10 k Da. Interestingly, observation of ethanol precipitation demonstrated that the ethanol precipitation has significantly algicidal activity. however, the water-layer did not inhibit the growth of the Skeletonema costatum.
