罗非鱼CD2BP2的克隆与对无乳链球菌的免疫应答

【摘要】 CD2BP2(CD2胞质结合蛋白2)是几个与CD2胞质区相互作用的蛋白之一,在CD2介导的T细胞激活以及核剪切中起到着重要的作用。在本研究中,尼罗罗非鱼(Oreochromis niloticus)的CD2BP2基因(On-CD2BP2)被成功克隆,序列分析表明On-CD2BP2具有已知CD2BP2的两个重要的结构特征,即GYF结构域和GPFXXXXMXXWXXXGYF基序,且其推测的基因结构也与已知的CD2BP2十分相似。通过荧光定量PCR检测On-CD2BP2在罗非鱼的m RNA表达量,结果表明,在健康的罗非鱼中,On-CD2BP2的转录本主要在头肾和脾脏被检测到;当用灭活的无乳链球菌对罗非鱼进行免疫刺激48 h后,On-CD2BP2在头肾、脾和脑中的表达表现出明显的时序表达模式,在脑的表达于24 h达到峰值,在头肾和脾的表达于48 h达到峰值。以上结果表明,On-CD2BP2在罗非鱼对细菌的免疫应答中起到着重要的作用。

【Abstract】 CD2BP2(CD2 cytoplasmic tail binding protein 2), one of several proteins interacting with the cytoplasmic tail of CD2, plays a crucial role in CD2-triggered T cell activation and nuclear splicing. In this paper, a CD2BP2 gene(On-CD2BP2) was cloned from Nile tilapia, Oreochromis niloticus. Sequence analysis showed that two important structural features, a GYF domain and a consensus motif GPFXXXXMXXWXXXGYF were detected in the deduced amino acid sequence of On-CD2BP2, and the deduced genomic structure of On-CD2BP2 was similar to the known CD2BP2. The m RNA expression of On-CD2BP2 in various tissues of Nile tilapia was analyzed by fluorescent quantitative real-time PCR. In healthy Nile tilapia, the On-CD2BP2 transcripts were mainly detected in the head kidney and spleen. While vaccinated with inactivated Streptococcus agalactiae, there was a clear time-dependent expression pattern of On-CD2BP2 after immunization and the expression reached the highest level at 24 h in the brain and 48 h in the head kidney and spleen. These findings indicated that On-CD2BP2 may play an important role in the immune response to bacteria in Nile tilapia.
