饲料中添加大黄素对团头鲂肝脏差异蛋白组的影响

【摘要】 本试验旨在研究饲料中添加不同水平大黄素(0、15、30、60、120 mg kg-1)对团头鲂肝脏抗氧化差异蛋白的影响。利用双向凝胶电泳(2-DE)技术分离对照组和最适添加量组(30mg kg-1)团头鲂肝脏差异蛋白,共获得差异蛋白点39个(表达丰度≥2倍),成功鉴定38个(上调8个,下调30个),共25种不同蛋白。通过构建网络互作模型,重点分析了载脂蛋白A-I(apo A-I)、果糖-二磷酸醛缩酶B(aldob)、HSP 70、谷胱甘肽过氧化物酶1(GPx 1)和谷胱甘肽S-转移酶(GST)等抗氧化相关差异蛋白。通过RT-PCR进一步对抗氧化相关差异蛋白转录水平表达趋势进行验证,结果表明,30mg kg-1添加组抗氧化相关基因的表达量最高且显著高于对照组(P<0.05),与蛋白水平表达趋势一致。因此,饲料中添加30mg kg-1大黄素能够提高团头鲂肝脏抗氧化相关蛋白的表达。
【Abstract】 The investigation was aimed to evaluate the change of hepatic antioxidant protein of dietary emodin supplementation(0、15、30、60、120 mg kg-1) in Megalobrama amblycephala. Two-dimensional gel electrophoresis(2-DE) was used to separate the hepatic differential protein of control and optimum emodin supplementation(30mg kg-1) group. 39 significantly differential expressed protein spots(up/downregulated≥2 fold) were separated, and in which 38 protein spots were successfully identified(8 upregulated and 30 down-regulated), including 25 unique proteins. By network interaction model, we mainly investigated the related antioxidant differential protein, including apolipoprotein A-I(apo A-I), fructose-bisphosphate aldolase B(aldob), HSP 70 protein, glutathione peroxidase 1(GPx 1) and glutathione S-transferase(GST). RT-PCR was carried out to compare the m RNA expression level of these antioxidant differential proteins of each group further. Results showed the 30 mg kg-1 supplementation group improved a significant m RNA expression of these antioxidant differential proteins compared with the control group(P < 0.05), which was consistent with the protein expression level. So the results suggested that 30 mg kg-1 emodin supplementation in feed can improve the expression of hepatic antioxidant protein in Megalobrama amblycephala.
