金钱鱼SF-1和dax1的分子特征及其在性逆转中的表达分析

【摘要】 为了解SF-1和dax1基因在金钱鱼性腺分化中的作用,本研究利用RACE技术克隆了金钱鱼(Scatophagus argus)SF-1和dax1基因c DNA全长序列,并研究17α-甲基睾丸酮(17α-methyltestosterone,MT)对该基因表达和性腺组织的影响。SF-1和dax1 c DNA全长分别为1708和1380bp,包括5′非翻译区(5′-UTR)分别为111和101bp、3′非翻译区(3′-UTR)分别为136和373bp,其开放阅读框(ORF)分别为1 461和906bp,分别编码486和301个氨基酸。组织分布结果显示dax1 m RNA在性腺、脾、脑及肠等组织中表达量较高,SF-1 m RNA在性腺和脾等组织中表达量较高。组织学结果表明,17α-甲基睾丸酮能有效诱导金钱鱼发生不同程度的性逆转,性腺中卵母细胞退化,精原细胞增殖。dax1m RNA表达量在MT处理40天后显著增加,而SF-1 m RNA在MT处理后其表达受到抑制。上述结果表明dax1和SF-1基因可能与金钱鱼性腺从雌性向雄性化发育有关,且dax1和SF-1之间存在一定的拮抗作用。

【Abstract】 The nuclear receptors Dax1 and SF-1 play important and antagonistic roles in gonad differentiation in mammals. To study their potential functions in teleost fish, the full length c DNAs of dax1 and SF-1 were cloned and characterized from spotted scat. Their expression patterns in adult tissues and juveniles treated with 17α-methyltestosterone(MT) and their gonada histological changes were investigated, respectively. The tissue specific expression results showed that dax1 was abundantly expressed in the gonads, spleen, brain and intestine. SF-1 was preferentially expressed in the gonad and spleen. Histological results showed sex reversal could be induced by MT, the degenerating oocytes and proliferations of spermatogonia were observed in gonad. At the beginning of sex reversal(40 days after treatment), the m RNA level of dax1 significantly increased, and the transcript level of SF-1 m RNA was inhibited at the point of 20 days following treatment with MT. Our results implied that dax1 and SF-1 might be involved in the gonad development from female to male and there was an antagonistic effect between dax1 and SF-1.
