氨茶碱对慢性阻塞性肺疾病小鼠Th1和Th17变化的影响

【摘要】 目的:探讨氨茶碱干预下对慢性阻塞性肺疾病(COPD)小鼠Th1和Th17变化的影响及其相关机制。方法:将30只Balb/c小鼠随机均分为对照组、模型组、氨茶碱处理组。采用烟熏法建立COPD模型小鼠。模型建立成功后,采用HE染色观察小鼠肺组织的病理形态学改变,采用酶联免疫吸附法(ELISA)检测小鼠支气管肺泡灌洗液(BALF)和肺组织匀浆的IFN-γ、IL-17变化。结果:与对照组相比,模型组小鼠的肺组织结构破坏,内有大量炎性细胞浸润,BALF和肺组织匀浆的IFN-γ、IL-17水平均升高;而氨茶碱处理组较模型组的组织形态学有所改善,BALF和肺组织匀浆的IFN-γ、IL-17水平均降低。结论:Th1、Th17失衡可能与COPD发病机制有关,氨茶碱可以改善COPD小鼠的炎症反应,并可能通过免疫调节对COPD小鼠起到治疗作用。

【Abstract】 Objective To investigate the effect of aminophylline on changes of Thl and Thl7 in mice of chronic obstructive pulmonary disease(COPD) and its mechanism.Methods Thirty Balb/c mice were randomly divided into control group,model group and aminophylline group.The COPD murine models were developed by smoke.After establishing models,the pathomorphology of murine lung tissues were observed by HE staining,and changes of IFN- 7,IL- 17 in murine Bronchoalveolar lavage fluid(BALF) and lung homogenate were determined by ELISA.Results The structure and Inflammation of lung tissues was more serious,the levels of IFN-γ,IL- 17 in BALF and lung homogenate were higher in model group than those in control group.The changes in model group could be reversed after treatment of aminophylline.Conclusion The nosogenesis was found related to cell Thl and Thl7 imbalance in COPD.Aminophylline could improve the inflammatory reaction,and may be therapeutic by Immune regulation in the COPD murine models.
