丹酚酸对氨氯地平抑制自发性高血压大鼠心肌肥大的增效作用研究

【摘要】 目的:通过比较丹酚酸联合氨氯地平及单独使用氨氯地平对自发性高血压大鼠(Spontaneously HypertensiveRats,SHR)大鼠心肌肥大的影响,探讨丹酚酸对氨氯地平的增效作用和作用机制。方法:将18只SHR平均分成3组,即模型组(蒸馏水10ml·kg-1·d-1)、氨氯地平组(氨氯地平1mg·kg-1·d-1)和联合给药组(丹酚酸浸膏粉100mg·kg-1·d-1+氨氯地平1mg·kg-1·d-1),另取SD大鼠作为正常对照组。连续给药8周后处死,检测各组大鼠心脏指数,左心室肥大指数。通过苏木精—伊红(hematoxylin-eosinstaining,HE)染色观察心肌组织基本形态;免疫组织化学法观察肿瘤坏死因子(tumor necrosis factorα,TNF-α)的表达水平;采用脱氧核苷酸末端转移酶介导的d UTP缺口末端标记法(Td T mediated d UTP nick end labeling,TUNEL)检测心肌细胞凋亡指数。结果:1联合给药组大鼠左心肥大指数较模型组明显下降(P<0.05),而氨氯地平组则仅有下降趋势而没有显著统计学差异(P>0.05)。2,联合给药组大鼠心肌组织TNF-α表达显著下降(P<0.05),氨氯地平组则未见改变(P>0.05)。3,联合给药组心肌细胞凋亡指数下降,与正常组接近(P>0.05),而氨氯地平组则未观察到这一现象。结论:丹酚酸能够增强氨氯地平抗心肌肥大的作用,两者联合使用能够降低心肌组织的炎症反应,抑制心肌细胞凋亡,预防高血压左室的形成。

【Abstract】 Objective:To explore the synergistic effect and mechanism of action of salvianolic acids on Amlodipine,by the way of comparing the effects of salvianolic acids combined with Amlodipine and Amlodipine longly in myocardial hypertrophy of SHR rats。Methods:18 SHR rats were randomly divided into3 groups:model group,amlodipine group and salvianolic acids combined with amlodipine group。In addition,SD rats were used as control group。The rats were contiously administrated for 8 weeks to test the index of rat’s hearts and left ventricular hypertrophy。HE staining method was used to observe basic shape of myocardium,immunohistochemistry method to observe the expressing level of TNF-α and Td T mediated d UTP nick end labeling( TUNEL) to detect apoptotic index of myocardial cells。Results:1,compared to model group,the index of left ventricular hypertrophy in combined administration group is significantly decreased( P < 0.05),but amlodipine group only tends to decrease( P > 0.05) without notably statistical differences( P > 0.05) 。2,the expressing of TNF-α in combined administration group is remarkably decreased( P < 0.05),however,amlodipine group has no change( P > 0.05) 。3,the apoptotic index of myocardial cells in combined administration group is similar to the control group( P > 0.05),but amlodipine group has no this phenomenon。Conclusion:salvianolic acids can increase the effect of amlodipine on resisting myocardial hypertrophy。The two combined used can decrease inflammatory reaction of myocardial tissue,inhibit apoptosis of myocardium cells and prevent the formation of left ventricular hypertrophy。
