草莓轻型黄边病毒抗血清的制备

【摘要】 草莓轻型黄边病毒(Strawberry mild yellow edge virus,SMYEV)是草莓生产中危害严重的一种病毒。本文探索在原核细胞中表达SMYEV的外壳蛋白(CP),并以其为抗原制备SMYEV抗血清的方法,建立利用ELISA检测SMYEV的技术体系。结果表明,SMYEV的CP基因在大肠杆菌中表达分子质量为52ku的融合蛋白。以纯化的重组蛋白为抗原,免疫新西兰兔,制备出针对SMYEV的抗血清,该抗血清稀释800倍后仍能有效检测草莓植株中的SMYEV。
【Abstract】 Strawberry mild yellow edge virus(SMYEV) is one of the most dangerous viruses affecting the strawberry production.In this study,we try to express the coat protein(CP) gene of SMYEV in prokaryotic cells,and to produce the antiserum against SMYEV with the recombinant CP as the antigen,thereby to develop the system of detection of SMYEV by ELISA.A 52 ku recombinant CP of SMYEV was expressed in Escherichia coli.After the purified recombinant protein was used to raise antiserum in the New Zealand rabbits,the antiserum was isolated and purified from the blood of rabbit and used to detect SMYEV in strawberry plants,which was still effective in detecting SMYEV by DAC-ELISA after diluted 800 folds.This study solved the problem that it is difficult to produce the antiserum against SMYEV with the virus particle as antigen.
