SFRP1 Reverses The Phenotype of Epithelial-Mesenchymal Transition in Human Lung Adenocarcinoma Cell Line A549

【摘要】 <正>Epithelial-mesenchymal transition(EMT) plays an important role in the pathogenesis of human non-small cell lung cancer(NSCLC).Wnt pathway is identified as a key regulator of normal tissue development,and its aberrant activation contributes to the process of EMT.Secreted frizzled-related protein 1(SFRPl),a Wnt signaling antagonist,is frequently downregulated in many tumors including lung cancer.However,whether SFRP1 is involved in EMT and tumor metastasis remains unclear.In this study,we constructed the transforming growth factorβ1(TGF-β1)-mediated EMT model in A549 cell line and observed that SFRP1 was downregulated in TGF-βl-induced EMT.Restoration of SFRP1 could reverse the phenotype of TGF-βl -induced EMT phenotype and inhibit the motility and invasiveness of A549 cell line.In addition,FH535,a revisable Wnt signaling inhibitor, exerted similar effects on TGF-βl-induced EMT phenotype.These results indicated that Wnt signaling played a critical role in TGF-β1-induced EMT,and SFRP1 was a potential biomarker for the treatment of NSCLC.
