酶解海洋低值鱼制备降血肽的研究

【摘要】 海洋低值鱼不仅含有丰富的蛋白质、维生素、比例合适的微量元素和牛磺酸,而且还含有海洋生物所特有的多种营养成分。研究发现海洋鱼类提取物具有增强免疫、抗疲劳、保护肝脏、防止高血压、抗肿瘤、抑制病毒等作用。因此,利用海洋低值鱼开发血管紧张素转化酶(ACE)抑制剂具有较大的潜力。本研究以海洋低值鱼为原料,以抑制率为指标,利用胰蛋白酶研究了酶解海洋低值鱼制备血管紧张素转化酶抑制肽的方法。同时,通过SephadexG-25凝胶进行柱层析对酶解产物进行分离,测定酶解物中血管紧张素转化酶抑制肽(降血压肽)的抑制率及其相对分子量的分布。研究结果表明:酶解液经层析分离,在第二个洗脱峰处得到的血管紧张素转化酶抑制肽抑制活性最高,其值为86.32%。本研究为新型降血压制剂或功能性食品的研制提供了依据。

【Abstract】 Marine low-value fish is not only rich in proteins,vitamins,proper proportion of trace elements and taurine,but also contains a variety of specific nutrients.Some studies have discovered that extracts of that marine fish can enhance immunity,anti-fatigue,protect the liver,prevent high blood pressure,antitumor and inhibit virus and so on.Therefore,low-value fish marine had a great potential to develop angiotensin-converting enzyme(ACE) inhibitors.In this study,marine low-value fish as raw material,method for preparation of angiotensin-converting enzyme inhibitor peptides was studied by using the inhibition rate as an indicator.Hydrolysates was separated by SephadexG-25 gel column chromatography to determine inhibition rate and molecular weight distribution of angiotensin-converting enzyme inhibitory peptides.The results showed that the highest activity value of 86.32%was obtained in the second peak of eluting.This study provided the basis for the new development of antihypertensive agent or a functional food.
