香豆素-酞菁锌轭合物的合成与离体抗癌活性研究

【摘要】 近年来,光动力治疗与其它治疗方法联合应用取得了较好的效果。本文选择具有较好抗癌活性及生物相容性的香豆素衍生物作为化学治疗结构单元,将其通过聚乙二醇链链接到具有高光动力活性的酞菁锌的母环结构上,获得系列香豆素-酞菁锌轭合物,期望在分子水平上获得化学与光动力联合治疗的新型药物。测定了轭合物光物理/光化学参数、细胞摄取量及离体光动力/化学灭活癌细胞能力数据,探讨了香豆素-酞菁锌轭合物离体抗癌活性的构效关系。
【Abstract】 In recent years,photodynamic therapy in combination with other treatment methods have achieved good results.In this paper,coumarin derivatives were employed as antitumor activity and biocompatibility chemotherapy structural unit,and zinc phthalocyanine was selected as the photosensitizing unit due to its high photodynamic activity,the two components were linked with a triethylene glycol chain.to give a series of coumarin – zinc(II) phthalocyanine conjugates.These conjugates are highly promising drugs for molecular-based dual chemo-and photodynamic therapy.Photophysical/photochemical parameters,in vitro cellular uptake and photodynamic/chemically inactivities toward cancer cells of these conjugates were determined,the structure-activity relationships of coumarin-zinc phthalocyanine conjugates were discussed.
