地塞米松抗坏血酸对牙髓细胞增殖分化的影响

【摘要】 目的:探讨地塞米松与抗坏血酸配伍对体外培养的人牙髓细胞增殖分化的影响。方法:组织块法体外培养人牙髓细胞,扩增后取第四代细胞随机分为两组,实验组牙髓细胞于基础培养液中加入10-8mol/L地塞米松及50μg/mL抗坏血酸培养,对照组单纯使用基础培养液培养。在细胞培养不同时间点分别检测细胞增殖、碱性磷酸酶活性、牙本质基质蛋白及牙本质涎磷蛋白基因表达及钙化结节形成情况。结果:实验组牙髓细胞增殖活性显著降低,与对照组相比有统计学差异(P<0.05)。碱性磷酸酶活性以及牙本质基质蛋白、牙本质涎磷蛋白基因表达水平则随着培养时间的延长而增高,实验组明显高于对照组(P<0.05)。实验组与对照组均有钙化结节形成,但是实验组钙化结节形成数量较多。结论:地塞米松抗坏血酸能显著抑制牙髓细胞的增殖活性,促进牙髓细胞的分化及矿化能力。

【Abstract】 Objective:To investigate the effects of dexamethasone and ascorbic acid on proliferation and differentiation of human dental pulp cells in vitro.Methods:Human dental pulp cells were isolated by explant culture methods and expanded to the fourth passage for experiments.The cells were randomly divided into two groups,experimental group and control group.The cells in the experimental group were treated with10-8 mol/L dexamethasone and 50μg/mL ascorbic acid in 2%FBS basal media.The cells in the control group were cultured in 2%FBS basal media only.At different time points,cell proliferation,alkaline phosphatase(ALP) activity,dentine matrix protein(DMP1) and dentine sialophosphoprotein(DSPP) gene expression,and calcified nodule formation were determined respectively.Results:Dexamethasone and ascorbic acid significantly inhibited cell proliferation of HDPCs compared with control group(P<0.05).Conversely,such combination significantly enhanced ALP activity of HDPCs(P<0.05) and promoted DMP1 and DSPP gene expression(P<0.05).Both experimental group and control group exhibited calcified nodules but experimental group showed more module formation than control group.Conclusions:Dexamethasone and ascorbic acid can significantly inhibit dental pulp cells proliferation activity,but stimulate the differentiation and mineralisation of dental pulp cells.
