超高压液相色谱串联质谱法同时测定硫色镰刀菌接种的马铃薯块茎中A型和B型单端孢霉烯族毒素

【摘要】 采用超高压液相色谱串联质谱法(UHPLC-MS/MS)同时测定硫色镰刀菌(Fusarium sulphureum)接种的马铃薯块茎中4种单端孢霉烯族毒素A型(T-2,DAS)和B型(3ADON,Fus-X)。线性回归系数R≥0.9996、回收率113.78-78.48%、精密度RSD≤5.37%、检出限(LOD)为2.0-5.0μg/kg(除了Fus-X 20.0μg/kg);检出量(LOQ)为5.0-15.0μg/kg(除了Fus-X 60.0μg/kg)。结果表明:该法适合于同时测定Fusarium sulphureum损伤接种的马铃薯块茎中T-2,DAS,3ADON和Fus-X四种单端孢霉烯族毒素的测定。另外,这4种单端孢霉烯族毒素不仅在马铃薯块茎病部被发现,而且在没有任何干腐病症状的健康部位也检测到该类毒素的存在。同时感病品种陇薯3号中这4种毒素的积累量远高于抗病品种陇薯6号中的积累量。

【Abstract】 A reliable and sensitive method for the rapid simultaneous determination of four Type-A(T-2,DAS) and type-B(3ADON,FUS-X) of trichothecenes was developed and successfully applied in potato tubers by UHPLC-MS/MS.The established methed was developed and further evaluated by determining the linearity(R≥0.9996),recovery(113.78-78.485),precision(RSD≤5.39),and sensitivity(LOD,2.0-5.0μg/kg,except for 20.0μg/kg of Fus-X;LOQ,5.0-15.0μg/kg,except for 60.0μg/kg of Fus- X).The method showed to be a suitable for simultaneous determination of T-2,DAS,3ADON,and FUS-X in inoculated potato tubers with F.sulphureum.In addition,it was found that T-2,DAS,3ADON,and Fus-X were detected predominantly in the lesion,and the toxin could also be determined in tubers without any disease symptoms.Meanwhile,the concentration of toxin in the susceptible cultivar(Longshu No.3) was significantly higher than in the resistant cultivar(Longshu No.6).
