早期反应基因IER5辐射诱导表达的转录因子研究

【摘要】 为弄清辐射诱导IER5基因的转录调控机制,采用生物信息学分析技术和缺失体构建、定点突变、ENSA及ChIP生物学实验方法,确定IER5基因启动子区域、转录因子和结合位点。实验发现IER5基因启动子最可能的较小范围为-408bp至-238bp。IER5基因具有两个转录因子,分别为GCF、NFI,其中GCF具有两个结合位点,分别位于在IER5基因启动子-388 bp~-382 bp和-274 bp~-270 bp处;NFI具有一个结合位点,该结合位点位于IER5基因启动子-362 bp~-357 bp。这两种转录因子对IER5基因的调控作用正相反,转录因子GCF对顺式作用元件起负性调节作用,并随照射剂量的增加而减弱;转录因子NFI对顺式作用元件起正性调节作用。

【Abstract】 In order to clarify regulation mechanism of IER5 induced by the radiation,This experiment used some bio-information methods and techniques to promoter region of IER5 gene and Binding site of its transcription factors.These experimental techniques included building deletant,site-specific mutagenesis,electrophoretic mobility shift assay(EMSA),chromatin immunoprecipitation(CHIP).The IER5 gene have two transcription factors, which respectively were NFI and GCF.GCF has two binding sites,which separately is located in-388 bp ~-382 bp and-274 bp~-270 bp in front of IER5 gene.NFI has a binding site,which located-362 bp~ -357 bp in front of IER5 gene.These two transcription factors played the completely contrary roles in gene expression of the IERS.The transcription factor GCF played negative regulation to IERS,and this action became lower with the increasing of radiating doses.On the contrary,The transcription factor NFI promoted the gene expression of IERS,and this regulation to IER5 might was associated with doses of radiation.
