福建部分茶树品种SSR遗传差异分析与指纹图谱建立
Analysis of Genetic Variance and Establishment of DNA Fingerprint of Several Tea Cultivars in Fujian

【摘要】 以课题组选育的10个茶树品种及福建5个参照品种为材料,利用SSR分子标记进行遗传差异分析并建立分子指纹图谱,旨在进一步明确这些品种相互之间的遗传差异,为开展茶树良种鉴定及知识产权保护提供参考。实验结果表明,筛选出的6对核心引物具有较高的多态性;不同核心引物之间的Nei’s基因多样性指数(H)和Shannon’s信息指数(I)的差异随参试材料数目的增加而减小;10个自育品种遗传多样性水平与全部15个参试材料相当,高于5个参照品种,但剔除早春毫、朝阳后,剩余8个品种遗传多样性水平低于5个参照品种;参试材料遗传相似系数变化范围为0.35~0.91,平均0.65,其中,朝阳与大红袍之间的遗传相似系数最小(0.35),紫牡丹与铁观音之间的遗传相似系数最大(0.91);选用W08、D7、L11引物组合建立了参试材料的分子指纹图谱,特征条带大小在210~630bp之间,可将参试材料逐一鉴别。
【Abstract】 Tea cultivar has been the most important factor in the production of tea, and played a key role in the development progress of tea industry in our country. In this paper, the genetic variance analysis and molecular fingerprint to 10 tea cultivars selected and cultivated by our research team and 5 other control cultivars in Fujian province were established by SSR molecular marker technology, aimed to further clarify the genetic differences of these cultivars and provide reference in identificating improved tea cultivars and protecting intellectual property rights. The results showed that, 6 core primer pairs with higher polymorphism were selected from published primer pairs. The variation of Nei ’s gene diversity index(H) and Shannon’s information index(I) to the different core primer pairs were decreased with the number of tested materials increased. The genetic diversity level of the 10 tea cultivars selected and cultivated by our research team was about the same with all the 15 tested materials, higher than the 5 other control cultivars, but the 8 tea cultivars, except Zao Chun Hao and Chao Yang of the 10 tea cultivars selected and cultivated by our research team, was lower than the 5 other control cultivars. The genetic similarity coefficient of the all tested cultivars ranged between 0.35~0.91 with an average of0.65, of which it was minimum between Chao Yang and Da Hong Pao(0.35) while maximum between Zi Mu Dan and Tie Guan Yin(0.91). The molecular fingerprint of the tested cultivars was established using the primer pairs combination of W08, D7, L11, and the characteristic stripes were between 210~630 bp, could identify the tested cultivars one by one.
