大鼠骨髓间充质干细胞向雪旺细胞诱导分化能力研究

【摘要】 研究骨髓间充质干细胞(BMSCs)向雪旺细胞诱导分化的能力,为组织工程神经选择一种理想的种子细胞。采用percoll密度梯度法分离骨髓间充质干细胞。流式细胞仪检测干细胞表面特异MARK CD29、CD45、CD80、CD90评价干细胞的生物学特性。采用化学诱导和共培养诱导两种方式诱导骨髓间充质干细胞向雪旺细胞分化,观察诱导后细胞的形态变化。采用免疫荧光染色检测雪旺细胞特异性标记物S-100和GFAP的表达,RT-PCR检测诱导前后雪旺细胞特异性标记物S-100、GFAP的表达。体外培养的大鼠BMSCs呈长梭形,CD29、CD90表达阳性,而CD45和CD80表达阴性,具有骨髓间充质干细胞的生物学特性。诱导后的细胞胞质向核收缩,呈典型的核周体形态,形成双极或多极状,细胞形态与雪旺细胞相似。免疫荧光染色S-100和GFAP表达阳性;RT-PCR显示诱导后的干细胞表达S-100和GFAP,而未诱导的不表达。体外分离培养的骨髓间充质干细胞能够向雪旺细胞诱导分化,诱导后其表型和分子特征与雪旺细胞相似,骨髓间充质干细胞是一种理想的组织工程神经的种子细胞。

【Abstract】 To investigate differentiation capability of bone marrow mesenchymal stem cells into Schwann cells and to select a idea.seed cells source for tissue engineering nerve.BMSCs were isolated with percoll density gradient centrifugation from rat bone marrow.Specific cell surface markers.of BMSCs were confirmed by flow cytometry toevaluate the biological characteristics.BMSCs were induced to Schwann cells by chemical-inducing and co-culture.The morphology was observed.Immunofluorescene staining and RT-PCR were used to identify S-100 and GFAP.The cultured BMSCs present spindle,BMSCs in this study were positive for CD29,CD90 and negative for CD80 and CD45.differentiation BMSCs is double pole spindle- like morphology similar to Schwann cells.Immunofluorescene and RT- PCR analysis revealed that the treated cells expressed the glial markers S-100 and glial fibrillary acidic protein indicative of differentiation.BMSCs can be induced to differentiate into Schwann cells in terms of morphologic features and phenotype and could be a idea seed cells for Tissue engineering nerve.
