血栓通对顺铂损伤HK-2细胞保护作用及其机制的研究

【摘要】 目的:研究血栓通(xueshuantong,XST)对顺铂损伤的人肾近曲小管上皮细胞(HK-2)增殖和相关指标的影响,探讨血栓通治疗顺铂肾损伤的可能机制,为防治顺铂肾损伤寻找新的途径。方法:体外培养HK-2细胞株,接种于96孔培养板:(1)培养72h后加入不同浓度的DDP,血栓通各剂量组在DDP作用12h后加入不同浓度XST,再培养48h后用MTT法检测细胞存活率。(2)建立DDP损伤HK-2细胞模型,12h后加入定量的XST。48h后测定细胞生长的存活率,检测细胞酶以及生化指标(SOD、GSH-PX活性和NO、MDA含量)。观察XST对损伤的HK-2细胞的保护作用。结果:(1)XST能升高HK-2细胞对DDP的IC50(半数抑制浓度)。(2)与对照组比较,DDP组细胞数量减少,形态不规则,各生化和酶学指标差异均有统计学意义;与DDP组比较,XST各剂量组细胞数量明显增加,形态规则,SOD、GSH-Px酶活性升高,MDA、NO水平降低,差异均有统计学意义(p<0.05或p<0.01)。结论:血栓通能促进HK-2细胞和DDP损伤HK-2细胞的增殖,改善DDP损伤HK-2细胞的生化指标和酶水平,对DDP损伤的HK-2细胞具有保护作用,其保护机制可能与其抗氧化作用有关。

【Abstract】 Objective: To study protective effects of Xueshuantong on renal function of Cisplatin-induced Nephroxicity in vitro.Exploring the possible mechanism of the Xueshuantong treatment and new ways for prevention and treatment of Cisplatin-induced Nephroxicity.Methods: Cultured HK-2cell lines were used to study the effects of Xueshuantong on Cisplatin-induced renal cell injury in vitro, HK-2cell were incubated in the presence of different concentrations of XST after culture for 72 h, The cell viability was detected with MTT method. Besides, the HK-2 injury model was established by Cisplatin (DDP) for 12h, then treated with a certain amount of XST. Value-added rate of cell growth, cell enzymes and biochemical indices (SOD,GSH-PX activity and NO,MDA content) were detected after 48h. Results: In vitro The IC50 of DDP-induced injury of HK-2 cells were increased by XST. Comparing with the DDP group, the cell number of XST treatment groups significantly increased, the morphology was ruled, the activity of SOD and GSH-Px increased, the levels of MDA, NO decreased, the difference was statistically significant(p<0.05 or 0.01). Conclusions: In vitro Xueshuantong can promote the proliferation of injuried HK-2 cells induced by DDP, improving the biochemical parameters and enzyme level. The results suggest that XST has a protective effect on HK-2 cells. The protective mechanisms of Xueshuantong may account for the influence on antioxidant effects.
