一测多评法测定枇杷叶有效部位中三萜酸成分的含量

【摘要】 目的建立枇杷叶总三萜酸有效部位中蔷薇酸、委陵菜酸、马斯里酸、科罗索酸、齐墩果酸和熊果酸的一测多评HPLC-ELSD含量测定方法。方法以熊果酸对照品为内参物,建立其与蔷薇酸、委陵菜酸、马斯里酸、科罗索酸、齐墩果酸的相对校正因子,采用校正因子计算这5种三萜酸成分的含量,实现一测多评;同时用外标法测定有效部位中该6种成分的含量,并比较计算值与实测值的差异,以验证一测多评的合理性、可行性和重复性。结果 15批有效部位中6种三萜酸成分含量的计算值与实测值无显著性差异。结论该方法操作简便,测定结果准确,实现了只用一个对照品同步测定枇杷叶总三萜酸有效部位中6个成分的含量,可为中药多指标成分质量评价模式提供参考。
【Abstract】 Objective This research aims to develop a quantitative analysis of multi-components by single-marker (QAMS) method for simultaneous determination of six triterpene acids in the effective fraction of Eriobotryae Folium. Method Ursolic acid was used as the internal reference substance, the relative correlation factors (RCF) of euscaphic acid, tormentic acid, maslinic acid, corosolic acid and oleanolic acid to ursolic acid were determined by HPLC-ELSD with good reproducibility. And then, the content of the five components was calculated according to the resulting correction factors, respectively. Results The contents of these six triterpene acids in 15 batches were determined by the external standard method. Rationality, feasibility and repeatability of the QAMS method was verified by comparing the results obtained from the two different methods. For the six triterpene acids, there was no significant difference between the quantitative results of the two different methods in 15 batches.Conclusion The method established in this research is accurate and feasible that it just need to assay single-marker (ursolic acid) for determination of six triterpene acids in the effective fraction of Eriobotryae Folium simultaneously. Therefore,this method can provide a new mind for the quality assess of traditional medicine.
