【摘要】 探索姜黄素(curcumin,cur)对血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)所致内皮细胞损伤模型的保护作用。用10-6mol·L-1AngⅡ构建内皮细胞损伤模型。Real time-PCR检测各组内皮细胞CD40、eNOS、DDAH1、DDAH2mRNA表达变化,并用Western Blot方法检测各组内皮细胞CD40表达变化情况,试剂盒测定细胞中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的活性以及细胞中丙二醛(MDA)的含量,活性氧(reactive oxygen species,ROS)检测试剂盒检测细胞内ROS水平。结果表明AngⅡ可显著上调内皮细胞CD40mRNA和蛋白的表达水平,同时伴有细胞内SOD、CAT活性下降,ROS含量明显增加,eNOS、DDAH1、DDAH2mRNA表达下降,姜黄素呈剂量依赖性地抑制AngⅡ所引起的CD40mRNA和蛋白表达的上调,减轻细胞内的氧化应激水平,并上调eNOS、DDAH1、DDAH2mRNA的表达水平。姜黄素能明显逆转AngⅡ造成的内皮细胞损伤,其机制可能与下调CD40、减轻细胞的氧化应激损伤有关。更多还原

【Abstract】 To investigate the protective effects of curcumin on endothelial cells damage induced by Angiotensin (AngⅡ). Endothelial cells were cultivated with 10 -6 mol·L -1 AngⅡ to establish damaged model. Real-time PCR was used to determine the expressions of CD40 mRNA, DDAH1 mRNA, and DDAH2 mRNA on endothelium cells. The protein expression of CD40 was detected by Western Blot analysis. The intracellular activity of SOD, CAT and MDA level were determined by corresponding detection kit, the lever of ROS production in endothelial cells was measured by ROS assay kit. AngⅡ increased both mRNA and protein level of CD40, down regulated the expression of eNOS mRNA, DDAH1 mRNA and DDAH2 mRNA, these observations were accompanied by decreased activity of SOD and CAT, increased intracellular level of MDA and ROS. Curcumin inhibited the expression of CD40 mRNA and CD40 protein induced by AngⅡ in a dose dependent way, and also alleviated the intracellular oxidative stress with increased mRNA level of eNOS, DDAH1, DDAH2. Curcumin reverse the endothelium damage induced by AngⅡ, CD40/CD40L pathway, oxidative stress are involved in the effect of curcumin.更多还原
