半滑舌鳎病毒性神经坏死病病原的研究

【摘要】 2012年8月,某养殖场半滑舌鳎鱼苗发生大规模死亡,死亡率高达100%。患病鱼苗呈现身体畸形弯曲、体色变深以及螺旋性游动的症状,组织病理检查发现病鱼脑部组织空泡化。采用浸泡感染的方式对25日龄和50日龄的半滑舌鳎鱼苗分别进行人工感染实验,结果发现25日龄的鱼苗在4天内累计死亡率达100%,而50日龄的鱼苗累计死亡率与对照组无显著差异。用LAMP和RT-PCR方法对患病半滑舌鳎鱼苗进行检测,结果呈神经坏死病毒阳性。用RT-PCR以及环化cDNA末端快速克隆(环化RACE)方法测定了该病毒的全基因组序列,结果表明,病毒RNA1由3104个核苷酸组成,RNA2由1433个核苷酸组成,系统发育树分析确定该病毒属于β-野田村病毒属,为赤点石斑鱼神经坏死病毒(RGNNV)基因型。本文通过临床症状、组织病理、人工感染试验和病毒序列分析研究,确认鱼类神经坏死病毒可以自然感染半滑舌鳎。
【Abstract】 The larvae and journals of half smooth tongue sole(Cynoglossus semilaevis) had outbreak disease with up to 100% mortality in hatchery in August 2012.The infected fish exhibit a bent body shape,dark body coloring and swimming in a whirling manner.Under light microscopy,the cell of brain showed conspicuous vacuolation.The half smooth tongue sole larvae of 25dph and 50dph were used for challenge experiment,the cumulative mortality of the larvae in 25dph was 100% in four days,but the cumulative mortality of the larvae in 50dph did not show diversity with the control.A positive of nervous necrosis virus was detected for infected half smooth tongue sole larvae by LAMP and RT-PCR.The complete sequence of virus was cloned by RT-PCR and the circular rapid-amplification of cDNA ends.The RNA1 of nervous necrosis virus was 3104bp,the RNA2 was 1433bp and the virus belonged to the member of RGNNV genotype in betanodavirues by phylogenetic analysis.This study determined the nervous necrosis virus of fish can nature infected the larvae and journals of half smooth tongue sole following clinical signs,histopathology,challenge experiment and genomics.
