免疫诱导模型大鼠肝组织病理变化的动态观察

【摘要】 目的观察异种蛋白诱导的肝纤维化模型大鼠肝组织的动态病理改变规律。方法雄性Wistar大鼠60只,随机分为2组,空白组8只不予任何处理;实验组52只在34天内皮下注射含4mg人白蛋白的不完全弗氏佐剂4次致敏,经间接ELISA法检测,淘汰阴性大鼠5只;余下47只大鼠分别于每周2和周5尾静脉注射白蛋白予以免疫攻击,第1周每只每次注射白蛋白2.5 mg,以后每周增加0.5 mg直至4.5mg,维持此剂量至第10周。从尾静脉免疫攻击开始每2周取大鼠6只,戊巴比妥麻醉后取肝组织,多聚甲醛固定后HE、Masson染色。至10周时按上述方法处理剩余全部9只及8只空白对照组大鼠(免疫攻击开始后14只大鼠死亡)。结果 HE染色显示,第2周时见部分汇管区内有少量炎性细胞浸润,偶见成纤维细胞或间质细胞增生;第4周时见汇管区炎性细胞浸润明显,可见胶原增生,偶见短纤维隔;第6周时见成纤维细胞增生,纤维细胞逐渐向汇管区和中央静脉延伸,胶原增生明显;第8周时见假小叶形成,小叶间隔胶原纤维沉积逐渐增多,偶见中央静脉纤维增生;第10周时假小叶形成,中央静脉纤维化增多。肝组织不同时间点的Ishak评分结果经配伍资料的秩和检验结果显示,和谐系数W=0.920,X~2=22.082,P<0.01,认为不同时间点Ishak病理评分不同;Masson染色显示,不同时间点组织Masson染色显示的胶原面积多样本配伍资料秩和检验,和谐系数W=0.987,X~2=23.679,P<0.01,可以认为不同时间点Masson染色中胶原纤维表达不同。结论随造模时间延长,第4周时出现纤维明显增生,并在第8周时开始出现假小叶。

【Abstract】 Objective To observe the time-dependent pathological changes law on heterologous protein-induced liver fibrosis model.Method 60 male rats are injected Freund’s incomplete adjuvant containing 4mg human albumin four times for sensitization in 34 days.The anti-human serum albumin antibody positive rats from these rats are choose to be injected human albumin 2.5mg each time twice a week,according to the increasing rate of 0.5mg each time to 4.5mg,maintenance the nose for 2.5 months.Four rats are chosen to anesthesia by chloral hydrate every 2 weeks and the liver tissues are obtained for the electron microscope observation and HE and Masson staining.Results HE staining showed that collagen deposition begins in 2th week,microstructure formed in 4th week,pseudolobuli formation increased,occasionally central vein fibrosis was seen in 8th week;common central vein fibrosis in 10th week.Rank test of Ishak score in different time points in liver tissues shows,W = 0.920,χ2 = 22.082,P <0.01,suggests that the Ishak pathological score at different times are different. Masson staining shows,multi-sample data rank test about collagen area at different times points result,W = 0.987,χ2= 23.679,P <0.01,it suggests that in different time points,collagenous fibers express different.Conclusion With the extended time,fiberoplasia is gradually significant in 4th week and the pseudolobule appears in 8th week.
