胃束带术治疗2型糖尿病模型大鼠的机制初探

【摘要】 目的链脲佐菌素(streptozotocin,STZ)结合高糖高脂饮食诱导建立2型糖尿病大鼠模型,观察胃束带术(gastric banding)的治疗效果,初步阐明其机制。方法:喂饲高糖高脂饲料4周后,用STZ 30 mg/kg体重一次性左下腹腔注射,建立2型糖尿病大鼠模型。将成模2型糖尿病大鼠分为手术组12只;假手术组12只;对照组10只。检测术前、术后1、2、3、4、8和16周空腹体重、血清葡萄糖(BG)、甘油三酯(TG)、总胆固醇(TC)、胰岛素(INS)等指标,进行统计分析。结果:手术组大鼠术前空腹体重为(387.2±37.0)g、BG值为(24.2±4.0)mmol/L、TG值为(4.3±0.5)mmol/L、TC值(2.4±0.6)mmol/L,术后均明显降低,术后16周,空腹体重为(324.4±22.5)g、BG值为(12.4±1.3)mmol/L、TG值为(2.3±0.4)mmol/L、TC值(1.4±0.3)mmol/L,与术前比较,均P<0.01。空腹血清胰岛素水平在观察期内无明显变化。结论:胃束带术能降低2型糖尿病大鼠的血糖及血脂,饮食控制体重减轻可能是胃束带术改善2型糖尿病大鼠模型血糖的主要机制。
【Abstract】 Objective To observe the effect of gastric banding on the rat model for type 2 Diabetes mellitus(T2DM ) induced by streptozotocin(STZ ) in combination with high-sugar and high-fat diets,and to primarily clarify its mechanism. Methods Rats were fed with high-sugar and high-fat diets for 4 weeks and injected STZ(30 mg / kg ) 4 weeks later.Type 2 diabetes rats were randomly allocated into operation with 12 rats;sham group with 12 rats and control group with 10rats.The fasting blood glucose(BG )、blood triglyceride(TC)、blood cholesterol(TC)、body weight,fasting insulin (INS ) were measured before and after operation 1st,2nd,3rd,4th,8th and 16th week.Results 16 weeks after operation,BG、TG,TC concentration were decreased,had statistical significance compares with the levels of control group P<0.01. Conclusion Gastric banding can reduce fasting BG、TC、TG in type 2 diabetes rats.Weight loss may be the major mechanism of blood glucose change in type 2 diabetes rats after gastric banding operation.
