应用傅立叶变换红外光谱技术对两株克雷伯氏菌快速分类

【摘要】 该文应用傅立叶变换红外光谱(FT-IR)技术对克雷伯氏菌属内肺炎克雷伯氏菌和产酸克雷伯式菌进行指纹图谱数据采集,建立了两株克雷伯氏菌的标准FT-IR光谱数据库。对光谱分别进行基线校正和归一化处理后,选取977.9~1805.3cm-1波数范围的特征指纹图谱区域,建立了基于主成分分析(PCA)和分级聚类分析(HCA)两种聚类分析模型。结果表明,两种聚类模型均可成功将两株克雷伯氏菌进行区分。傅立叶变换红外光谱分析方法简便、快速、易操作,结果重现性好,是一种区分肺炎克雷伯氏菌和产酸克雷伯式菌的有效方法。
【Abstract】 Fourier transform infrared(FT-IR) spectroscopy combined with multi-variate statistics,has been a novel tool to differentiate and identify microorganisms in recent years.In this study,FT IR absorption spectra(4 000-600 cm-1) of two strains of Klebsiella,viz.,Klebsiella pneumoniae and Klebsiella oxytoca,were collected,and a standard spectral library was created.After treatments of automatic baseline correction and normalization,FT-IR fingerprint absorption spectra over the range of 977.9-1 805.3 cm-1 were analyzed by chemometric methods.Two cluster models based on principal component analysis (PCA) and hierarchical cluster analysis(HCA) were established.The results showed that two strains of Klebsiella could be well differentiated,and the standard spectral library created could be used to compare with those of target Klebsiella and identify them.As a rapid,easy-to-use and accurate technique,FT-IR spectroscopy is an effective tool to differentiate and identify two strains of Klebsiella.
