中国野生垂穗披碱草种质的SRAP和RAPD分析

【摘要】 对来自我国青藏高原和新疆地区的64份垂穗披碱草(Elymus nutans Griseb.)材料用SRAP和RAPD标记进行遗传多样性和亲缘关系的分析,并测定垂穗披碱草的遗传变异和各地理类群的遗传多样性水平。结果表明:2种标记都显示出供试材料具有较高的遗传多样性水平(PPB=85.86%,90.39%),且新疆地区材料的遗传多样性水平高于青藏高原地区;2种标记具有显著的相关性(r=0.986,P<0.01),2个标记相结合而得的聚类图表明相似生态地理环境的材料可以聚为一类;分子方差分析(AMOVA)揭示了地理类群内部和地理类群间分别有44.82%和55.18%的变异;青藏高原和新疆地区的材料存在明显的遗传分化,气候和山脉等生态地理条件以及繁育系统等可能是使材料发生遗传变异的重要因素。这些结果将为垂穗披碱草种质的育种以及种质资源的收集和保存提供依据。

【Abstract】 The diversity and relationships among 64 E.nutans accessions of Qinghai-Tibetan Plateau and Xinjiang were analyzed using sequence-related amplified polymorphism(SRAP) and random amplified polymorphic DNA(RAPD) molecular markers in order to measure the genetic variability and compare the level of diversity.The results showed that:High genetic diversity was found using two molecular markers in E.nutans(PPB=85.86%,90.39%),Xinjiang region had higher level of diversity than Qinghai-Tibetan Plateau; two markers had a significant correlation(r = 0.986,P<0.01),the dendrogram of two markers showed that the accessions from the similar origin frequently clustered into one group;the analysis of molecular variance (AMOVA) showed that the proportion of variance within and among geographical groups was 44.82%and 55.18%,respectively;the distinct genetic differentiation was revealed between the accessions of Qinghai-Tibetan Plateau and Xinjiang.The ecogeographical conditions such as climate and mountain ranges and mating system could be driving factors for genetic variation.These results will facilitate the breeding program and germplasm collection and conservation of E.nutans.
