夹角余弦相似度对色谱指纹图谱共有峰和非共有峰灵敏度的评价

【摘要】 色谱指纹图谱相似度是评价中药质量稳定性的有效手段之一,其中向量夹角余弦相似度在药材质量稳定性评价中应用广泛。但是,当指纹图谱峰面积分布范围宽时,夹角余弦对数据的差异响应不灵敏。本研究以土茯苓色谱指纹图谱数据为实例研究了夹角余弦相似度对色谱指纹图谱的共有峰和非共有峰响应的灵敏程度,结果表明当待测样本与参照样本峰比例差异较大时,夹角余弦相似度对色谱指纹图谱的共有峰和非其有峰响应的灵敏度相差很大。因此,在应用夹角余弦相似度进行原料药质量稳定性评价时,建议色谱指纹图谱的非共有峰以不同于共有峰的权重纳入相似度的计算,使得相似度的计算结果能更好地评价原料药质量的稳定性。

【Abstract】 The similarity of Chromatographic Fingerprints is one of the effective approaches evaluating the stability of Chinese Medicine,and cosine of the angle plays an important role in the application of similarity. However,cosine of the angle is insensitive to the data difference when the distribution range of the data sets is large. The sensitivity of cosine of the angle to common peaks area and non-common peaks area was investigated by the experiment data of chromatographic fingerprints of Smilax glabra Roxb.in this study,which result showed that the sensitivity of cosine of the angle to common peaks area and non-common peaks area was related to chromatographic fingerprints data of 9 different samples,and the weight coefficient of non-common peaks should be determined by chromatographic data of different samples.Therefore,It was suggested to take non-common peaks area into the similarity calculations in different weighs from the weigh of common peaks area,which is a promising attempt to better evaluate the quality stability of Chinese crude drug.
