饮用水中4-枯基酚检测方法及其急性毒性和遗传毒性研究

【摘要】 目的饮用水中4-枯基酚定性定量检测方法建立及4-枯基酚的急性毒性和遗传毒性研究。方法采用固相萃取-气相色谱质谱联用(GC-MS)法建立水中4-枯基酚检测方法。以寇氏法研究4-枯基酚对昆明小鼠的急性毒性(经口);以Ames试验、小鼠骨髓嗜多染红细胞微核试验评价遗传毒性。结果 GC-MS方法检出限0.02μg/L,加标回收率81.0%~86.2%,相对标准偏差6.8%~14.8%。4-枯基酚对昆明小鼠经口LD50为1141.5mg/kg(95%可信区间:744.75-1749.62mg/kg);各剂量组(0.005-0.1mg/皿)TA97、TA98、TA100及TA102菌株回变菌落数与自发回变组接近;不同剂量染毒组(67.5mg/kg/d、135mg/kg/d和270mg/kg/d)小鼠骨髓嗜多染红细胞微核率1.2‰-1.6‰,与对照组相比无显著差异(P>0.05)。结论所建方法灵敏度高、选择性好,适于饮用水中4-枯基酚定性定量分析。4-枯基酚为无毒物质,无致突变性及非染色体着丝粒阻断剂。
【Abstract】 Objective To establish the qualitative and quantitative detection method for 4-cumyphenol in drinking water and study its acute toxicity and genotoxicity.Methods The solid phase extraction-gas chromatography/mass spectrometry(GC/MS) method was used for the establishment of detection method for 4-cumyphenol in water.The Kunming mice were employed for testing the oral acute toxicity of 4-cumyphenol and its genotoxicity was assessed through Ames test and micronucleus assay in mouse bone marrow polychromatic erythrocytes.Results The detection limit,the recovery rate,and the relative standard deviation(RSD) of the detection method were separately 0.02μg/L,81.0%-86.2%and 6.8%-14.8%.The oral LD50(95%confidence interval) for Kuming mice of 4-cumyphenol was 1141.5mg/kg(744.75-1749.62mg/kg).The reverse mutation of Salmonella typhimurium in various dose groups(0.005-0.1mg/plate) of TA97, TA98,TA100 and TA102 strains was similar to that in control group.The micronucleus rates of mouse bone marrow polychromatic erythrocytes in different dose groups(67.5mg/kg/d、135mg/kg/d and 270mg/kg/d) were 1.2‰-1.6‰,which showed no significant difference comparing with the control group(P>0.05).Conclusions The established detection method with high sensitivity and satisfying selectivity are suitable to qualitative and quantitative determination of 4-cumyphenol in drinking water.4-Cumylphenol is the no toxic compound,with no mutagenicity and is not a broken agent of centric.
