心衰康颗粒对心梗大鼠心肌局部白介素-6、白介素-1β表达的影响

【摘要】 目的:通过研究心衰康颗粒对心梗后大鼠心肌局部白介素-6(IL-6)和白介素-1β(IL-1β)表达的影响,阐释该方剂干预左室重构的作用机理。方法:结扎Wistar大鼠左冠状动脉根部建立心肌梗死动物模型,随机分为假手术组、模型组、心衰康大剂量组、心衰康小剂量组和卡托普利对照组,药物干预8周,采用免疫组化法观察心肌局部IL-6和IL-1β的含量。结果:所有给药组IL-6和IL-1β的表达与模型组比较均不同程度的下降;且大剂量组与西药组作用相似(P>0.05),与小剂量组比较有显著差异(P<0.01或<0.05)。结论:心衰康颗粒能降低心肌IL-6和IL-1β水平,此应为干预实验性心肌梗死大鼠左室重构的机制之一。

【Abstract】 Objective:To study the effects of Xinshuaikang granule (XSK) on the expression of myocardial interleukin-6(IL-6) and interleukin-1β(IL-1β) in rats with myocardial infraction(MI) and elaborate its mechanism underlying the intervention of left ventricular remodeling.Methods:We established acute myocardial infraction(AMI) in Wistar rats by the ligation of the left coronary artery.The rats were divided randomly into five groups: sham-operated group,MI group,high dose of XSK treated group,low dose of XSK treated group and captopril treated group.After 8-week administration of medicine,the immunohistochemistry was adopted to observe the expression of myocardial IL-6 and IL-1β.Results:The expression of myocardial IL-6 and IL-1βin all medicine treated groups declined to some degree compared with MI group.The decline of IL-6 and IL-1βwas similar between high dose of XSK treated group and captopril treated group(P>0.05),however,the expression of IL-6 and IL-1βin both of the two groups was different with low dose of XSK treated group(P<0.01或<0.05).Conclusion:XSK granule could reduce myocardial IL-6 and IL-1βlevels significantly,which may be one of the mechanisms underlying the inhibition of left ventricular remodeling in MI rats.
