Regulation of Wnt receptor activation

【摘要】 <正>Wnt signaling plays important roles during embryogenesis,tissue homeostasis as well as tumorigenesis.Frizzled and LRP5/6 are the two major cell surface receptors required for the canonical Wnt signaling.While the mechanism of Frizzled activation is largely unknown,extensive progress have been achieved in the recent years on the understanding of LRP5/6 activation upon Wnt stimulation.Phosphorylation of the intracellular domain of the LRP5/6 receptor is believed to be the main event for receptor activation and several kinases have been identified.In addition,receptor oligomerization and acidification of the microenvironment have also been described which further complex this process.We have identified a novel cell membrane protein which is able to enhance LRP6 phosphorylation and do so in a relatively distinct manner.I will present evidence to show that this protein likely functions as a membrane scaffold for Wnt receptor activation.
