MPVA软件设计及其在乳房炎易感性多座位聚合效应分析中的应用

【摘要】 【目的】以奶牛隐性乳房炎为例来介绍MPVA软件在多座位聚合效应分析中的应用。主要具体介绍MPVA软件的原理、算法及操作。【方法】以南方某大型奶牛场634头(其中102头患乳房炎)中国荷斯坦牛为试验材料,采用PCR-SSCP、直接测序法分析CXCR1-5′端和编码区及IL8 2789/2862编码区两个基因共7个SNP位点遗传多态性,并用MPVA法分析CXCR1和IL8基因各位点突变及其基因型组合对奶牛乳房炎易感性的影响。【结果】MPVA分析发现CXCR1-1830,CXCR1-1768和IL8 2789/2862三位点交互作用对奶牛乳房炎易感性达到显著水平。【结论】CXCR1-1830,CXCR1-1768和IL8 2789/2862三位点交互模型作为奶牛乳房炎易感性相关的基因联合选择模型最佳,MPVA可用于奶牛乳房炎易感性多基因分析模型。
【Abstract】 【Objective】The purpose of this article is to introduce the use of MPVA software in analyzing pyramiding effects.we just take Chinese Holstein for example,then we introduce the principle, algorithm and operation of the software.【Method】The polymorphis-ms of 7 SNP sites including the 5’ flanking region and coding region of CXCR1 and exon 4 and exon 5 of IL8 were detected by PCR-SSCP and direct sequencing for 634 Chinese Holstein,including 102 individual mastitic cow,from one northern farm of China,and to identify the interaction of CXCR1 and IL8 and tⅡeir impacts on the risk of mastitis suscepti-bility using Multi-Locus penetrance variance analysis(MPVA).【Results】The interactions of CXCR1-1830-1768-EL8 2789/2862 showed significant impacts on the mastitis susceptibility of cow using MPVA model.【Conclusion】The interactions of CXCR1-1830-1768-IL8 2789/2862 was the best multi-genotype model to estimate the risk of mastitis susceptibility of Chinese Holstein.MPVA can be used to estimate the risk of mastitis susceptibility of Chinese Holstein.
